Introdution

• The posterior capsular opacification (PCO) remains a frequent complication of cataract surgery ranging from 3.5% to 55.6%, depending on the design and material of the intraocular lens.

• One of this treatment the polymerization of the acrylic hydrophobic surface IOLs with polyethylene glycol. This kind of treatment has shown a significant reduction in protein deposition and cell adhesion in the IOL (Lee et al., 2007)

Purpose

Methods

• The research was Approval by the Botucatu Medical School

Ethics committee.

• We performed the epithelial cells culture from the anterior

capsule of 144 patients undergoing cataract extraction by undergoing cataract extraction by

Methods

• 3,000 cells of each culture was seed on the Hydrophilic IOLs (Ioflex®, Mediphacos, Belo Horizonte - Brazil) untreated and treated with polyethylene glycol, both the samples in triplicate.

• The treated IOLs were submitted to treatment with polyethyleneglycol (PEG 300) 392°F evaporated to a glass chamber at a pressure of – 0.0045 Torr, using radio frequency energy 5 to 15W for 30 minutes.

Methods

• IOLs were left at 98,6° F in a humidified incubator with 5% CO2 for 2 hours for cell adhesion to occur .

• Then nutrient with 20% fetal bovine serum was added, and the cultures kept in the incubator for 24 hours.

Methods

• 126 (87.5%) cultures out of 144, showed the rounded pattern, and 18 (12.5%) the dendritic pattern.

Results

• Overall, there was less cell adhesion on IOLs treated compared to untreated controls, with statistical significance (p <0.05).

Results

• By using primary cell cultures, it was possible to add a new variable (cell type) to the experiment, which allowed us to get closer to the real situation. The finding of two distinct types of cells was not expected

• Analysis of the two cell pattern revealed their distinct behavior. Although no statistical difference was found, the dendritic pattern seems to be more adherent in comparison with the round pattern, and the adhesion inhibition in the treated IOLs group was higher in the dendritic pattern in comparison with this control group.

Discussion

• These findings lead us to suppose that patients with dendritic pattern cells in culture, were more likely to develop PCO because this kind of cells could be transformed into activated myofibroblasts.

• We could not find in the literature experiments that cite and compare these two types of cells in culture, since only cell

strainsstrains were used for this kind of study.
