ACROSS RESEARCH STRUCTURE


Advancesinseparationsciencehaveprovidedtheimpetusforexcitingnewdevelopmentsinthebiologicalsciences(eg.genomics,proteomicsandmedicine),pharmaceuticalsciences(eg.drugdiscoveryandcharacterisation),environmentalsciences(eg.ultra-traceresidueanalysis),forensicscience(eg.illicitdrugs,DNAfingerprinting,andexplosivesresidues)andotherareas.Thediscoveryofnewmodesofseparationscienceinvolvinganalysis,characterisationandpurificationwillbeessentialtothesefields.Separationsciencealsobridgesthenanoscalethroughtothemacroscale,withcommonelementsoftheoryandimplementation.AdvancesinseparationsciencewillthereforebeanimportantdriverbehindaverybroadspectrumofAustralianscience,rangingfromnewdevelopmentsinnanotechnologytonovelbiomaterials.Itsimportanceasanenablingsciencecannotbeoverstated.

The Australian Centre for Research on Separation Science (ACROSS)wasestablishedin2001asastrategicagreementbetweenkeyresearchersattheUniversityofTasmaniaandRMITUniversity(withUniversityofWesternSydneyjoiningACROSSin2008)toformaconsortiumofprominentAustralianresearchersinseparationscience.Thisconsortiumwassupportedfinanciallybytheparticipatinginstitutionstopursuethefollowingaims:

(i) maintainanoutstandinglevelofinternationalrenowninresearchonseparationscience,

(ii) coalesceandenhanceAustralianresearchonseparationscienceintoanorganisedstructureoperatingwithacoordinatedresearchplanwhichaddressesandexploitsthemostexcitingandinnovativethemesinmodernseparationscience,

(iii) provideenablingresearchandresearchtrainingofthehighestqualitywhichsupportsandadvancesallmajorareasofAustralianscience.

Australian research in separation sciencehaslongenjoyedanexcellentinternationalreputation,earnedbytheindividualactivitiesoftalentedresearchers.ACROSSoffersanorganisationalandresourcebasethroughwhichtheseindividualresearcherscanworkinacoordinatedandsynergisticmannerunderaseriesofstructuredandinterlockingresearchprograms.Thisavoidsduplicationofeffort,allowsresourcesandexpertisetobesharedandvalue-addedopportunitiestobeprovidedbroadlytoindustry,academiaandtheNation,andalsoestablishesmuchneedednationaltrainingfacilitiesinseparationscience.

Research in ACROSShasbeenestablishedusingfocusedresearchthemestoprovidebothfundamentalandappliedresearchoutcomesinseparationscience.ACROSSdrawstogethermulti-site,internationallyprominentandgenuinelycollaborativeresearchteams,havingcomplementaryskillsandsynergisticresource-baseexpertise,andcommittedtofocusedprogramsofnationalsignificance.Theresearchstructurelistedbelowgroupsresearchthemesusingthethreemajorphasesofaseparationandalsoshowsthemajoroutcomeareasinwhichthesethemesarebeingapplied.

Separation science involves the study of fundamental processes and materials for the separation and subsequent measurement of specific molecules, usually when these are present in very complex mixtures. It finds use in all of the chemical and biological sciences and in many areas of engineering.

Staffing changes2010sawtheappointmentatUTASofProfessorMirekMackaandProfessorPavelNesterenkoasNewStarsProfessors,DrDarioArruaasapostdoctoralfellowwhoisworkingonamonolithcharacterisationprojectfundedbyanARCDiscoveryGrantandDrViktorDrganasapostdoctoralfellowworkingonanionchromatographyoptimisationprojectfundedbyanARCDiscoveryGrant.ProfessorPhilipMarriottmovedfromRMITtoMonashUniversity.VisitorsspendingsignificantperiodsoftimeinACROSSin2010includedProfessorBrettPaullandDrEkatarinaNesterenko(Ireland),andHernanCortes(USA).Inaddition,therewereseveralinternationalPhDstudentswhoconductedpartoftheirresearchprojectsinACROSS.

Research outcomesACROSShasundertakenanextensiveprogramoffundamentalandappliedresearch,withnumerousindividualresearchprojectsbeingundertakenin2010.Theseresearchtopicscanbefoundelsewhereinthisreport.Somenotablefeaturesofthisresearcharethecontinuedstrongemphasisonmonolithicstationaryphases,thedevelopmentofnewhyphenatedandmultidimensionalseparations,continuedresearchintothesearchforbiomarkersfortheTasmanianDevilFacialTissueDisease,studiesonstationaryphaseselectivityandcapacity,retentionmodellinginionchromatographyusingcomplexelutionprofiles,andanextensiveprogramontheapplicationofseparationscienceasatoolforcounter-terrorism.

FundingFundingforACROSSin2010hastotalled$3,488,747,with$2,284,645(66%)comingintheformof14highlycompetitivegrantsfromtheAustralianResearchCouncil(ARC)[1FederationFellowship,2FutureFellowships,1QEIIFellowship,6DiscoveryGrants,2LinkageGrants,2LinkageInfrastructureandFacilitiesGrants].TwonewARCFellowshipswereawardedin2010forcommencementin2011.Additionalfinancialsupportwasprovidedbyarangeofgovernmentbodiesandindustries,withmajorcontributorsbeingPfizer(throughthePfizerAnalyticalResearchCentre)andtheNationalSecurity,ScienceandTechnologyBranch(AustralianFederalGovernment)forcounter-terrorismresearch.

Achievements in 2010ACROSSstaffcontinuetofeatureprominentlyontheinternationalseparationsciencescene.Membersofstaffheldthreeeditorshipsofinternationaljournalsin2010andalsoappearedontheeditorialboardsof18otherjournalsofanalyticalchemistryorseparationscience.SpeakersfromACROSSmadeinvitedpresentationsatmostofthemajorinternationalconferencesandsymposiaonseparationscience.

AnumberofothernotableachievementsbyACROSSstaffoccurredin2010.DrRobertShelliewastherecipientofaTasmanianYoungTallPoppyAward.DrGregDicinoski,DrLitoQuirino,DrEmilyHilderandDrMichaelBreadmorewereallpromotedtoLevelD.DrEmilyHilderandDrRobertShelliereceivedUTASVice-Chancellor’sAwardsforResearchExcellence.DrRobertShellieandDrRosanneGuijtreceivedUTASRisingStarAwards.DrRobertShelliereceivedtheRACIAnalyticalandEnvironmentalDivisionCattrallMedal.TheACROSS-CE-ScansystemforexplosivedetectionwasafinalistintheTasmanianStateGovernmentC-StarAwards.DrGregDicinoskireceivedtheawardforbestoralpresentationattheAustraliaandNewZealandForensicScienceSymposium.DrAshrafGhanemreceivedanEndeavourAwardfromtheAustralianGovernment.Finally,ProfessorPaulHaddadreceivedtheinauguralUTASVice-Chancellors’MedalforResearchExcellenceandhewasalsotherecipientofthe2010RACIAppliedResearchAward.

Pfizer Analytical Research Centre (PARC)Pfizer,theworld’slargestresearch-basedpharmaceuticalcompany,establishedin2007anewcollaborativeresearchcentreentitledthe“PfizerAnalyticalResearchCentre”(PARC),atTheUniversityofTasmania,Hobart,Tasmania.ResearchinPARCtargetsinnovationandenhancedproductivityinthepharmaceuticalanalyticalsciences.Theaimhasbeentoproduceadvancementsintheanalysisofpharmaceuticalsthroughhigh-throughput,faster,andsmarteranalyticalsystems,therebyallowingtheseproductstobebroughttothemarketplaceearlierandinamorecosteffectiveand‘green’manner.

ThePARCmultidisciplinarycollaborationcreatesacentreofexcellenceforthedevelopmentofnovelpharmaceuticalanalyticalmethodsbycombiningtheexpertiseofACROSSwiththatofworld-leadingpharmaceuticalscientistsfromPfizer.Thismulti-milliondollarcentrebringseconomicandfinancialgaintoboththeUniversityofTasmaniaandtotheStateofTasmania.Pfizer,througha$3.5minvestment,hasfundedtheappointmentofresearchstaffandstudents,purchaseofequipmentandtheprovisionofrunningcostsforthevariousprojects.TheUniversityofTasmaniahascontributed$850kfortheconstructionofthestate-of-the-art,world-standard,purpose-builtlaboratorycomprising450m2ofinstrumentalbenchandofficespace,inwhichPARCishoused.TheTasmanianStateGovernment,throughtheDepartmentofEconomicDevelopmentandTourism,hascontributed$267kforscholarshipstofacilitatetherecruitmentofinternationalPhDstudents.ThiscontributionhasenabledACROSStorecruithighcalibrestudentsfromfourdifferentcountriesthroughouttheworld.

In2010thecentrecomprised11staffandstudents,includingscientistsandtechnicalemployees.ThesepersonnelincludeaDirector,DeputyDirector,3post-doctoralfellows,5researchhigherdegree(PhD)students,andatechnicalofficer,withadditionalcontributionsfromcurrentACROSSstaff.EachPfizerprojectinvolvesclosecollaborationbetweenACROSSresearchersandPfizerscientistslocatedintheUKoratvarioussiteswithintheUSA.

Preparative-scale Capillary Multidimensional Gas Chromatography for Absolute Identification of Volatile Compounds

TheRMIT/Monashgrouphassuccessfullydemonstrateduseofapreparativescalemulti-dimensionalcapillaryGCsystemforprecisefractioncollectionofsingleGCpeaksfromcomplexmatrices,tosupportimprovedchemicalcharacterisationofcompounds.ThesinglebiggestproblemarisingfromtheuseofcapillaryGCresideswithuncertaintyofidentificationofseparatedcompounds.Thishasbeenlargelyaddressedinthepastbytheadventofmassspectrometrymethodsfordetectionofresolvedpeaks.However,thereisstillconsiderableresidualuncertaintyinidentificationandallocationofstructuralformulaeduetoimprecisionoflibraryspectralmatchingandinterpretation.Thisisrecognisedinanumberofimportantareas.Forinstance,essentialoilcomponentshavemanymassspectralionsincommonacrosstheterpenestructures,soMSfeaturesforminorcomponentsleadstouncertaintyinMSassignments.Thisisalsocommonlyfoundforisomersofmanyothercompounds.

Inordertoaddressthisgenerallimitation,wefirstrecognisedthatMSisabottlenecktoadequatecharacterisation.Therearemanysolutionstomolecularcharacterisation,usuallyrequiringalternativespectroscopicmethods,buttheseareoftennotreadilyhyphenatedwithGC.Second,ifwewanttoprovideaspectroscopicsolution,thenweneedtoisolatethecompoundfromthematrix(spectroscopyworksbestonpurecompounds),collectit,andthenperformdifferentmethodsofanalysis.Fortunately,onceacompoundisisolated,thenmanydifferentmethodscanbeusedtocharacterisethecomponent.Finally,weneedtoaccomplishthecompleteisolationofthecompound,andforthiswewanttoaddresspotentiallyverycomplexmixtures.Multidimensionalcapillarygaschromatography(MDGC)isthemostappropriate

technologyforthis.Toscale-upthepreparativeprocess,itisnecessarytoinjectmultiplequantitiesofsample,andtoalsocollectthesinglecompoundmanytimes.Wedevelopedafullyautomatedmethodforunattendedoperationofthetrappingsystem,capableofcollectingintheorderof100µgquantitiesofcompound,whichissufficientfor1HNMRand2DNMR.ThisallowsustoaddNMRtotheMSresultsoftheisolatedcompounds.Indeed,collectionofmuchsmalleramountscanalsoallowsatisfactoryidentification,dependingonspectralcomplexityandsensitivityofthespectroscopicmethod.

Ourstudiesfirstappliedthemethodtogeraniolinacomplexessentialoilsample,andthentocharacterisationofmethylnaphthaleneisomersinacrudeoilsample.ThecomparativeNMRresultforasimpleorganic–1,4-dimethoxybenzene–demonstratedthatthiscouldbeagoodinternalstandardforourapproach.Itispossibletouseasmallvariantofthemethodtocollectmultipleisolatedcompoundsfromacomplexsample,andtherebycreatenewmultiple-componentmixturesfromnaturalmaterials.Flow-switchingcapabilitiesthatsupportsuchprocessesarereadilyimplemented.Finally,ithasbeendemonstratedthatnotonlycanNMRbeusedtocharacterisethematerialsfromacatalystproductthatwasdifficulttoseparatebyclassicalliquidchromatographymeans,butsuitablecollectionandcarefulcrystallisationallowstheminutequantitiesofcollectedpurecomponentstobeabsolutelycharacterisedbyuseofX-raycrystallography.Suchacapabilityhasbeenrarelyreportedintheliterature,andthescopefordiscoveryofnewinformationbasedinthisapproachisournexttask.

A New Design in Chromatography Columns

Continuingfromworkinitiatedin2008withThermoFisherScientificasresearchpartner,thedesignofanewconceptchromatographycolumnhasbeentakentothepointof‘ProofofPrinciple’in2010bytheUWSgroup.Thebirthofthe‘activeflowmanagement’columnwasrealised.Inthe‘activeflowmanagement’projecttwonewtypesofchromatographycolumnsweredesigned,bothofwhichcouldbeemployedatthepreparativeandanalyticalscales.

Thefirstnewdesigninthechromatographycolumnwastheparallelsegmentedflowcolumn.Thesecondnewdesignwasthecurtainflowcolumn,whichalsoincorporatedaspectsofparallelsegmentedflow.Inbothdesignsaspecialfittingandfritareattachedtothecolumnoutlet.Thisallowsforthesolventandsampletobeselectivelyisolatedfromthebulkflowofmobilephaseatthecolumnoutlet.Inthecurtainflowdesign,aspecialinletheaderisattachedtothecolumn,whichalsocontainsaspeciallydesignedinletfrit.Thisfritandheaderallowsthesampletobeintroducedtothecolumninthecentralregionofthebed,andacurtainflowofmobilephaseisolatesthesamplefromthewallregionofthecolumn.Theparallelsegmentedoutletfittingensuresthatasthesampleisdetectedelutionmaintainstheadvantagesofthe‘infinitediameter’columnsincetheparallelsegmentedflowendfittingseffectivelymimictheperformanceofanendcolumndetector,ratherthanatraditionalbulkdetector.Whiletherehavebeennumerousattemptstobuildthe‘infinitediameter’

columnovertheyearsthisisthefirstcolumnofitskindtobedesignedthatcanbeusedwitharegularautoinjectorandemployaregularbulkdetector.Usingthesenewcolumnstheefficiency,asmeasuredbythenumberoftheoreticalplates,increasedbyasmuchas50%,anddetectionsensitivityincreasedby150%atthepreparativescaleandmorethan50%attheanalyticalscale.

Perhapsthemostsignificantadvantageofthecolumnsisthatthevolumeofanelutingsamplebandcanbecontrolled,withexperimentationshowingthatan85%reductioninpeakvolumemaybefeasibleforthesamelevelofdetectionasaregularcolumn.Thisisimportantbecauseitmaymeanthatveryhighthroughputseparationscanbeachievedusinghyphenatedtechniques,wherethedetectorissensitivetothevolumeload,suchasinLC-MS.ThesecolumnsshouldthereforeexperiencewidespreadapplicationinallfacetsofcolumnliquidchromatographyespeciallyinLC-MS.

In2010twonewPhDstudentsjoinedthegroupatUWS:MichelleCamenzuli,whohasbeeninvolvedinthestudyonthedesignofthenewconceptHPLCcolumn,andMarkTrudgett,whoisstudyingadvancedaspectsofmultidimensionalHPLC.Hisprojectspecificallystudiesseparationsinthetimeandfrequencydomains,pavingthewayforveryfastseparationsofcomplexsamples.
