Since opening its doors in 2015, the Beijing Advanced Innovation Center

for Structural Biology (ICSB) at Tsinghua University has quickly gained

a reputation for producing prolific amounts of high-quality research in

the life sciences.

In four years, the center has published 50 high-impact research

papers in Nature, Science, and Cell, and helped define and develop this

rapidly developing field in China, says Yigong Shi, director of the ICSB.

The former dean of the School of Life Sciences at Tsinghua University

dramatically advanced the field of molecular biology with his ground-

breaking discoveries about spliceosomes, the cellular machines that

make all of the proteins our bodies need to function. Shi explains that

the senior faculty at the center are training the next generation of

exceptional scientists while simultaneously producing world-leading

basic and translational research.

“The institute put itself on the map when my team published

the first near-atomic-resolution structure of the spliceosome. Some

scientists have called this discovery one of the holy grails of structural

biology,” says Shi, who won China’s foremost science award, the Future

Science Prize, in 2017. “Our research improved our understanding of basic

biological processes and should help to identify new drug targets in

the future.”

Dreaming of breakthroughsWhen Shi returned from studying in the

United States in 2007, he began to

envision a new center based in

Beijing that could conduct

advanced research of

structural biology. Over

the following decade, Shi

encouraged

Chinese

scientists to

join him at

Tsinghua,

and those 15 scientists helped him lay the intellectual foundations for

ICSB. Now, the center has 23 independent laboratories that house more

than 40 postdoctoral fellows and 200 graduate students. The young and

creative team has an average age of 40 years.

Shi’s mission is to ensure that ICSB produces the next generation of

top-level molecular biologists who can integrate traditional scientific

approaches with new technologies and research methods. In order

to ensure that the pipeline of talent never runs dry, and cultivate the

talents of early-career structural biologists, the center has established

a training program. The Advanced Innovation Fellow program selects

trainees to work on risky, difficult research and engineering projects.

After completing this two-year program, the most outstanding

researchers are given the opportunity to join the center as senior

researchers.

“We have recruited more than 20 postdocs in our program,” says Shi.

“Some of them have made significant achievements and shown their

great potentials in structural biology.”

For example, Xuerui Wan, who entered the program in 2018, was

elected as one of the four winners of the Science & SciLifeLab Prize for

Young Scientists in the same year.

“She is a rising star in structural biology of the spliceosome, and her

research has given important insights into the mechanism of pre-RNA

splicing,” says Shi.

A collaborative approachThe field of structural biology demands a cross-disciplinary approach,

and the ICSB has poured resources into creating a team of scientists, from

areas such as computing, electronics, mechanical engineering, chemistry,

physics, maths, and biology, that can work together with biophysicists,

structural biologists, and cell biologists to work on key problems and

produce major breakthroughs.

Hongwei Wang, who earned his undergraduate degree and Ph.D. at

Tsinghua University, researches cryo-electron microscopy (cryo-EM)—a

tool that can help researchers examine the structures of macromolecules

at extremely high resolutions. According to Wang, one of the greatest

challenges in his field of research is to build a team with a wide enough

disciplinary background to handle the challenges posed by the projects.

“I have built strong collaborations with scientists in nano-material

science and scientists in precision instrument implementation to tackle

the major technical challenges, such as cryo-EM specimen preparation

and cryo-EM structural analysis throughput,” he says.

Most recently, Wang and his team helped set a world record for the

smallest protein (52 kDa streptavidin) to be solved at atomic resolution

using single-particle cryo-EM reconstruction. Using cryo-EM, he and his

team hope to solve a wide range of drug target macromolecules

in their relative native states without crystallization, which

could lead to more efficient drug discoveries.

Designs on healthWhile researchers at the ICSB are devoted to unraveling the mysteries

of life at an atomic level, they are also keen to see their work solve real-

life problems in the future.

Haitao Li researches the biological mechanisms that switch genes

on or off at the chromosomal level—a field called epigenetics—and

explores how drugs can be developed to treat medical conditions, such as

cancer, heart diseases, and diabetes, which are influenced by epigenetic

dysfunction.

For example, his laboratory has recently identified how the YEATS

domain proteins relate to the development of leukemia and developed

compounds that can be used in ENL gene-targeted therapy. Li works

closely with hospitals across China and thinks there will be many

opportunities to translate his basic research into applications for the

benefit of patients, given the strong support he receives from the

ICSB.

Scientists at the center benefit from state-of-the-art facilities, such

as high-end cryo-EM machines, and proteomic and imaging core facilities,

he says. In addition, there is an ethos of intellectual curiosity that enables

groundbreaking work.

“The mission of the center is to resolve the mystery of biological

structures in diverse functional systems of life,” he says. “We are given

the freedom to choose our projects, aim high, and think deeply.”

Xinquan Wang, a biophysicist, agrees that these factors will help

scientists at ICSB translate research that can benefit the health of

Chinese citizens. His group is researching the structure of the interleukin 1

family of cell-surface receptors. Wang hopes their study will enable the

development of antibodies that are able to modulate the activities of

these receptors, which are linked to inflammation in the body.

He uses cutting-edge X-ray crystallography and cryo-EM tools in his

work, but adds that he and his team need to combine structural biology

methods with other functional techniques to gain more insights into the

scientific questions.

“Collaboration is also important for our study, especially for applying

the knowledge from our research to the development of agents with

potential therapeutic applications,” he says.

Looking to the futureShi has his sights set on new avenues of scientific enquiry that will

open up possibilities for cellular research at ICSB and Tsinghua University.

For instance, he is interested in how biological systems interact with

electromagnetic waves.

“On the pragmatic side, the applications of this kind of research

could be enormous,” he says. “For example, if electromagnetic waves of a

longer wavelength can induce a specific interaction in cells, which leads

to cell death in cancer cells, then that could be used as a therapy to treat

cancer patients.”

The prize-winning scientist highlights that there are many forces

interacting with our bodies that are unknown to us.

“There are many things we don’t know because we don’t see them.

Using state-of-the-art tools, we can gain a glimpse into this process,”

he says. “To me, this is an enormously interesting area that has not been

explored by many scientists.”

An international outlook

As the ICSB enters its fifth year, its international reputation continues

to grow.

Since its establishment, the ICSB has welcomed scientists from all

over the world, including Nobel Prize–winning chemist Brian Kobilka,

says Shi. Such collaborations ensure that the center will continue to grow

as an international hub for world-leading research in the life sciences.
