Study on Volatile Compounds in Fig Fruits

Abstract Odor is an important quality index for fresh fruit consumption. Odor of the fruit is associated with volatile
compounds. The volatiles in three cultivars of fig fruits (Black Genoa, Brown Turkey and Kadota) were determined by solid-phase microextraction/gas chromatography-mass spectrometry. The results indicated that the volatile compounds in figs could be classified into nine groups: hydrocarbons, ketones, alcohols, aldehydes, esters, acids, ethers, phenols and pyrans. All volatile compound groups were found in Black Genoa and Brown Turkey, while pyrans were not found in Kadota. Black Genoa had the highest amount of volatiles (3,135 ng/g D.W.) followed by Brown Turkey and Kadota, respectively. The majority of these volatile compounds were hydrocarbons and aldehydes. Based on the odor activity value, most of the potent aroma volatiles of figs were aldehydes. (E)-2-hexenal (green, fruity and fresh odor) was the odorant compound with the highest concentration in all the three cultivars of figs. Keywords: fig fruits, volatile compounds, cultivar
บทคดยอ กล�นเปนดชนคณภาพท�สาคญในการบรโภคผลไมสด กล�นของผลไมมความสมพนธกบสารระเหย (volatile
compounds) จากการศกษาชนดและปรมาณสารระเหยของผลมะเด�อฝร�ง 3 พนธ คอ แบลกจนว บราวนตรก และคาโดตา ดวยวธ solid-phase microextraction/gas chromatography-mass spectrometry พบวาสารระเหยในผลมะเด�อฝร�งสามารถแบงไดเปน 9 กลม ไดแก ไฮโดรคารบอน คโตน แอลกอฮอล อลดไฮด เอสเทอร กรด อเทอร ฟนอล และไพแรน โดยสายพนธแบลกจนวและบราวนตรกมสารระเหยครบท 2ง 9 กลม ในขณะท�สายพนธคาโดตาไมมสารระเหยกลมไพแรน สารระเหยของผลมะเด�อฝร�งสายพนธแบลกจนวมปรมาณมากท�สด (3,135 นาโนกรมตอกรมน 2าหนกแหง) รองลงมา ไดแก สายพนธ บราวนตรกและคาโดตา ตามลาดบ สารระเหยสวนใหญของผลมะเด�อฝร�งเปนสารระเหยในกลมไฮโดรคารบอนและอลดไฮด เม�อพจารณาจากคา odor activity value สารระเหยท�มแนวโนมเปนสารใหกล�นสาคญของผลมะเด�อฝร�งสวนใหญเปน สารระเหยในกลมอลดไฮด โดยผลมะเด�อฝร�งท 2ง 3 พนธมปรมาณ (E)-2-hexenal มากท�สด ซ�งเปนสารระเหยท�ใหลกษณะ กล�นเขยว (green) กล�นผลไม (fruity) และกล�นสด (fresh) คาสาคญ: มะเด�อฝร�ง สารระเหย สายพนธ

คานา มะเด�อฝร�ง (Ficus carica L.) เปนไมผลจดอยในวงศ Moraceae เชนเดยวกบหมอน เปนผลไมเพ�อสขภาพท�นยม
บรโภคสด ปจจยท�มผลตอคณภาพของผลไมมหลายประการเชน ลกษณะปรากฏ ส เน 2อสมผส คณคาทางโภชนาการและ กล�นรส นอกจากการเปนแหลงของสารประกอบฟนอลกซ�งมคณสมบตเปนสารตานออกซเดชนแลวอกปจจยท�มความสาคญคอคณภาพดานกล�นรสของมะเด�อฝร�งซ�งเม�อมะเด�อฝร�งสกจะมกล�นหอม โดยกล�นของผลไมสดจะถกกาหนดโดยสารใหกล�นท� สาคญ (odor-active compounds) ปจจบนการศกษาสารใหกล�นท�สาคญตอกล�นของผลไมไดรบความสนใจเพ�มมากข 2นเน�องจากเปนลกษณะเฉพาะของผลไมแตละชนด ดงน 2นงานวจยน 2จงมงศกษาชนดและปรมาณสารระเหยของผลมะเด�อฝร�งสด เพ�อใหไดองคความรพ 2นฐานของผลมะเด�อฝร�งสดสายพนธตางๆ ท�ปลกในประเทศไทยและเปนขอมลสาหรบการพฒนาในการแปรรปดานตางๆ เพ�อใหไดผลตภณฑท�มความเหมาะสมในการนาไปใชประโยชนตอไป
อปกรณและวธการ 1. การเตรยมตวอยาง นาผลมะเด�อฝร�ง 3 สายพนธ ไดแก สายพนธแบลกจนว คาโดตา (จาก จ.เชยงใหม) และบราวนตรก (จาก จ.พจตร)
เกบเก�ยวชวงเดอนมกราคม-กมภาพนธ ลางทาความสะอาด ผ�งใหแหง ห�นใหมขนาด 1x1x1 เซนตเมตร และแชไนโตรเจนเหลวทนท จากน 2นนาไปบดดวยเคร�องบดตวอยาง (Waring blender) (ทาการทดลองท�คณะอตสาหกรรมเกษตร ม.เกษตรศาสตร)
2. การสกดสารระเหย ทาการสกดสารระเหยดวยวธ solid-phase microextraction (SPME) ( ดดแปลงจาก Oliveira et al., 2010a) ท�
อณหภม 45 องศาเซลเซยส นาน 15 นาท ป�นดวยความเรว 200 รอบ/นาท จากน 2นทาการ adsorption สารระเหยเหนอตวอยางดวยไฟเบอรชนด PDMS-DVB ขนาด 65 ไมโครเมตร ท�อณหภม 45 องศาเซลเซยส เปนเวลา 30 นาทแลวนาไปทาการ desorption ดวยความรอนท� injection port ของเคร�อง GC อณหภม 250 องศาเซลเซยส เปนเวลา 5 นาท
3. การวเคราะหชนดและปรมาณสารระเหย นาตวอยางท�ไดจากการสกดในขอ 2 มาวเคราะหดวย gas chromatography-mass spectrometry โดยใชสภาวะ
เคร�องแบบ splitless mode แยกสารระเหยดวยแคปลารคอลมน 2 ชนด คอ คอลมน HP-5 และคอลมน FFAP ตามสภาวะของ Ware et al. (1993) บงบอกชนดของสารระเหยโดยแมสสเปกโตรมเตอร การระบชนดสารระเหยโดยการเปรยบเทยบขอมล mass spectrum ของสารแตละชนดกบฐานขอมล Wiley 275 library คานวณปรมาณความเขมขนของสารระเหยโดยการหาพ 2นท�ใตพคของสาร แตละชนดเปรยบเทยบกบพ 2นท�ใตพคของสารมาตรฐาน ( -2 undecanone) ท�ทราบความเขมขนแนนอน แลวรายงานเปนความเขมขนสมพทธของสารชนดน 2นๆ )นาโนกรมตอกรมน 2าหนกแหง ( สาหรบการพจารณาสารระเหยสาคญของผลมะเด�อฝร�ง พจารณาจากคา odor activity value (OAV) ซ�งคานวณไดจากความเขมขนสมพทธของสารระเหยแตละชนดและคาความเขมขนต�าสดท�สามารถรบรได (threshold) ของสารระเหยชนดน 2นๆ ดงสมการท� 1 โดยสารระเหยท�มคา OAV มากกวา 1 มแนวโนมวาเปนสารระเหยสาคญในการเกดกล�น (Grosch, 1993)
4. การประเมนผลทางสถต วเคราะหขอมลดวยโปรแกรมสาเรจรปทางสถต โดยใชวธ Analysis of Variance (ANOVA) เพ�อวเคราะหความ
แปรปรวน และเปรยบเทยบคาเฉล�ยโดย Duncan’s Multiple Range Test (DMRT) ท�ระดบความเช�อม�นรอยละ 95
OAV = (1) ความเขมขนของสารระเหยในตวอยาง
threshold ของสารระเหยใหกล�นชนดน �นๆ

ผล สารระเหยท�พบในผลมะเด�อฝร�งแบงเปน 9 กลม ไดแก ไฮโดรคารบอน คโตน แอลกอฮอล อลดไฮด เอสเทอร กรด
อเทอร ฟนอล และไพแรน โดยผลมะเด�อฝร�งสายพนธแบลกจนวมปรมาณสารระเหยมากท�สด (3,135 นาโนกรมตอกรมน 2าหนกแหง) รองลงมาไดแกสายพนธบราวนตรก และคาโดตา ตามลาดบ ซ�งสารระเหยสวนใหญของผลมะเด�อฝร�งเปนสารระเหยกลมไฮโดรคารบอน และอลดไฮด ในขณะท�สายพนธคาโดตามปรมาณสารระเหยกลมเอสเทอรมากกวาสายพนธอ�นๆ (Figure 1) สาหรบสารระเหยท�มแนวโนมเปนสารใหกล�นสาคญของผลมะเด�อฝร�งสวนใหญเปนสารระเหยกลมอลดไฮด (Table 1) ซ�ง (E)-2-hexenal มปรมาณมากท�สดในทกสายพนธ แตถาพจารณาคา OAV พบวา (E,E)-2,4-nonadienal มคา OAV มากท�สดเม�อเปรยบเทยบกบสารระเหยชนดอ�น โดย (E,E)-2,4-nonadienal พบเฉพาะในสายพนธแบลกจนวและ บราวนตรก สาหรบ (E)-2-decenal มแนวโนมเปนสารระเหยใหกล�นสาคญท�พบเฉพาะในสายพนธบราวนตรก สารระเหยกลมไฮโดรคารบอนท�มแนวโนมเปนสารใหกล�นสาคญไดแก l-limonene และ β-caryophyllene สาหรบสารระเหยท�มแนวโนมเปนสารใหกล�นสาคญกลมแอลกอฮอลไดแก 1-octen-3-ol และ l-linalool โดยสายพนธแบลกจนวและคาโดตามปรมาณ 1-octen-3-ol ไมแตกตางกน (p>0.05) ในขณะท�สายพนธบราวนตรกมปรมาณ l-linalool มากท�สด (p<0.05) สารระเหยท�มแนวโนมเปนสารใหกล�นสาคญกลมเอสเทอรสวนใหญพบในผลมะเด�อฝร�งสายพนธคาโดตา สวนสารระเหยท�มแนวโนมเปนสารใหกล�นสาคญกลมอเทอร (1,8-cineole) ในผลมะเด�อฝร�งท 2ง 3 สายพนธมปรมาณไมแตกตางกน (p>0.05) สาหรบ guaiacol สารระเหยในกลมฟนอลน 2นไมพบในผลมะเด�อฝร�งสายพนธแบลกจนว
วจารณผล
จากการทดลองพบวาผลมะเด�อฝร�งแตละสายพนธมชนดและปรมาณสารระเหยแตละกลมแตกตางกน ท 2งน 2เน�องจาก สายพนธท�ตางกนจะมสารประกอบทางเคมท�ทาหนาท�ในการแสดงกล�นรสของผลไมแตกตางกน (Christensen et al., 2007) สาหรบสารระเหยกลมไฮโดรคารบอนซ�งพบปรมาณมากในผลมะเด�อฝร�งแตไมมแนวโนมเปนสารใหกล�นสาคญ เน�องจากสารระเหยกลมน 2มคา threshold คอนขางสง (Baltes and Song, 1994) สาหรบสารระเหยกลมอลดไฮด เชน (E)-2-hexenal เปนสารระเหยท�ใหลกษณะกล�นเขยว (green) กล�นผลไม (fruity) และกล�นสด (fresh) (Leffingwell, 2004) สวน (E)-2-decenal ซ�งพบเฉพาะในสายพนธบราวนตรกน 2นสอดคลองกบรายงานของ Buttery et al. (1981) ซ�งพบสารน 2ในผลมะเด�อฝร�งสายพนธ Calimyrna โดยสารน 2ใหลกษณะกล�นเขยว (green) และกล�นไขมน (fatty) ) Leffingwell, 2004) สาหรบ l-limonene เปนสารระเหยใหลกษณะกล�นสด (fresh) กล�นหวาน (sweet) และกล�นพชตระกลสม (orange citrus) สวน β-caryophyllene ใหลกษณะกล�นไม (woody) และกล�นเคร�องเทศ (spicy) ซ�งสารระเหยท 2งสองชนดน 2สอดคลองกบรายงานของ Oliveira et al. (2010b) ซ�งพบในผลมะเด�อฝร�งสายพนธ Pingo de Mel, Branca Tradicional, Borrasota Tradicional, Verbera Preta และ Preta Tradicional สาหรบ 1-octen-3-ol เปนสารระเหยท�ใหลกษณะกล�นเหด (Genovese et al., 2007) และ l-linalool เปนสารระเหยท�ใหลกษณะกล�นสม (citrus) กล�นดอกไม (floral) และกล�นหวาน (sweet) (Leffingwell, 2004) ethyl hexanoate ซ�งเปนสารระเหยท�มปรมาณมากท�สดในกลมเอสเทอร มรายงานวาเปนสารระเหยใหกล�นสาคญในแอปเป2ลพนธฟจ (Song, 2010) 1,8-cineole เปนสารระเหยใหกล�นสาคญของพชสมนไพร เชน กะเพรา (Barbieri et al., 2004) โดยสารน 2ใหลกษณะกล�นการบร (camphoraceous) กล�นเยน (cool) และกล�นสดช�น (fresh) (Leffingwell, 2004) สาหรบ guaiacol เปนสารท�ใหลกษณะกล�นหวาน (sweet) และกล�นคลายควน (smoke-like)(Leffingwell, 2004)
สรป
สารระเหยท�พบในผลมะเด�อฝร�งสวนใหญเปนสารระเหยกลมไฮโดรคารบอน และอลดไฮด สาหรบสารระเหยท�มแนวโนมเปนสารใหกล�นสาคญสวนใหญคอสารระเหยกลมอลดไฮดซ�งใหลกษณะกล�นเขยว กล�นผลไม และกล�นสด โดยผลมะเด�อฝร�งสดจะมชนดและปรมาณสารระเหยแตกตางกนไปตามสายพนธ สาหรบผลมะเด�อฝร�งสายพนธแบลกจนวมปรมาณ β-caryophyllene มากท�สด ในขณะท�สายพนธบราวนตรกและคาโดตามปรมาณ (E)-2-hexenal มากท�สด จากผลการทดลองทาใหไดขอมลพ 2นฐานดานสารระเหยของผลมะเด�อฝร�งท�ปลกในประเทศไทย เพ�อเปนขอมลสาหรบการพฒนาในดานตางๆตอไป
