2 **ระบบภมคมกน (immune system)

แอนตเจน (antigen; Ag) หรอ อมมโนเจน (immunogen) มความเหมอนหรอแตกตางกนอยางไร?

แอนตเจน คอ สารใดๆ กตามทสามารถจบกบโมเลกลของแอนตบอด (antibody) หรอตวจบจ าเพาะของ T cell (T cell receptor) ได

เรยกคณสมบตนวา “antigenicity” อมมโนเจน คอ สารใดๆ กตามทสามารถกระตนใหเกดการตอบสนองทางภมคมกนแบบ adaptive immunityได

เรยกคณสมบตนวา “immunogenicity”

**All immunogens are antigens, but not all antigens are immunogens!!

คณสมบตของ immunogen ทด 1. Foreigness เปนสงแปลกปลอมของรางกาย sequestered antigens ใน immunological privileged sites เชน เชออสจ

(sperm), กระจกตา (cornea) และเนอสมอง ?

2. Molecular size สารทมขนาดโมเลกลใหญ ( >10 kDa) จะม immunogenicity ดกวาสารทมขนาดเลก

Toll-like receptor s(TLRs) are an evolutionarily conserved family of pattern

recognition receptors expressed on many cell types. play essential roles in innate immune responses to

microbes. Toll was originally identified as a Drosophila gene

involved in establishing the dorsal-ventral axis during embryogenesis of the fly.

Subsequently, it was discovered that the Toll protein also

mediated antimicrobial responses.

Cell-mediated immunity (CMI)->อาศยเซลล การท างานของ CMI จะเกยวของกบ T cells เปนหลก Subpopulation ของ T cells แบงตามหนาทไดดงน 1. T helper cells (Th cells) ->ชวยกระตน HMI และ CMI 2. Cytotoxic T cells (Tc cells or CTLs) -> ฆาเซลลทตดเชอ ไวรสและเซลลมะเรง 3. Regulatory T cells (Treg cells) -> ควบคมการท างาน ของ Th cells และ CTLs T cells สามารถแบงออกเปน 2 ชนด ตามโปรตนทอยบนผวเซลล ไดแก CD4+ T cells และ CD8+ T cells

3. ปฏกรยาลบลางฤทธ (Neutralization) สารพษหรอเอนไซมตางๆ จะถกลบลางฤทธ (neutralized) เมอท าปฏกรยากบแอนตบอด

• แอนตบอดทลบลางพษ toxin เรยกวา antitoxin • แอนตบอดตอไวรสลบลางฤทธในการตดเชอไวรสเรยก neutralizing antibody (NT-Ab)

5. ปฏกรยาระหวางแอนตเจนกบแอนตบอดทมการตดฉลาก (label) สามารถตดฉลากไดทงแอนตเจนและแอนตบอดสวนใหญนยมตดฉลากแอนตบอดมากกวา สารทนยมใชตดฉลากไดแก เอนไซม เชน horseradish peroxidase, alkaline phosphatase, glucose oxidase สฟลออเรสซน (fluorescein dye) เชน fluorescein isothioicyanate (FITC) สารกมมนตภาพรงส เชน I125 , 3H (tritium)
