Chemistry

Ionic liquids are salts that are liquid attemperatures below 100 degrees; some areliquid even at room temperature. Just like tablesalt, they consist of positively and negativelycharged ions, but with ionic liquids these arelarger and more complex. As the crystalstructure shatters, the salt becomes liquid - andan ionic liquid is the result. Properties of ionicliquids can be managed using combinations ofanions and cations that are accurately matchedto each other. This tailor-made approach resultsin liquids with, for example, very good solventproperties, thermal and electro-chemicalstability or reduced risk of fire and explosion.In addition, most of these substances are moreviscous than well-known traditional solvents.This is in stark contrast to many of the commonsolvents required in organic synthesis, whichare often toxic, very volatile and highlyflammable. The cost to the user and theenvironment is considerable, and the risk tosafety from explosions significant.

Using ionic liquids as solvents avoids theseissues. Potential applications are almostlimitless: "In the lab we often have to carry outaqueous conversions and extractions.As ionic liquids form phases with many organiccompounds this is one good area to use them,but the possibilities are vast." said Dr. Urs Welz-Biermann from Merck.

Various applications

Ionic liquids are also very good for use inenzyme reactions. Enzyme catalysts (andcatalysts in general) can be significantly moreefficient using ionic liquids, with increasedyields and ease of separation. This provides apositive economic benefit too as the catalystdissolved in salt can easily be separated outand fed back into the reaction.

A new standard for the coulometric determination of water in oil samples

The Merck MAS-100® Microbial Air Monitoring Systems are amongst the most accurate examples ofthe Anderson impaction principle of sample collection. Air is aspirated through a perforated lid with400 holes, impacting growth media in a standard 90 mm petri dish or 60 mm contact plate.A state-of-the-art mass flow sensor assures a constant air flow rate (real time) of 100 L/mincontinuously regulating the flow during sample collection and automatically adjusting for variationsin media fill levels and individual perforated lids as well as air density.

'Close-it' control seals

For the closure of sacks, cartons etc. thathave been pierced by zone samplers.In contrast to normal labels, 'Close it' selfadhesive seals adhere well to sacks thatare coated with powder (plaster, flour etc).The aluminium backing is super strong andthe seal can be labelled with appropriateinformation with a pen, pencil or felt pen.

The POLYTRON® PT 3100 Quattro in combinationwith the famous "W" shredder design is anindispensable tool that you can rely on.The preparation of hard and brittle, sometimestough and fibrous samples is often difficult andtime-consuming work. Deep-frozen cereal grain,or, for example, broad beans are extremely hardand abrasive; the connective tissue from meat isvery fibrous and tough. One disadvantage ofstandard dispersing instruments is that hardparticles get caught in the teeth of the stator,and long-fibrous tissues catch on the teeth of therotor. This stops processing and consumes thesample whilst leaving the tool virtuallyimpossible to clean.

The "W" design of the POLYTRONPT3100 Quattro generator has nosurfaces with parallel faces; theteeth get thinner towards theend. Coarser particles are,therefore, not jammed in thestator teeth but thrown outwardsby the turbulence of the rotorand thus reprocessed. In order tominimise abrasion caused by hardparticles, the rotor and stator ofthe dispersing tool are coatedand hard metal plain bearingsare used in the generator.

These modifications increase the life spanconsiderably.

A large amount of mechanical force is requiredfor efficient preparation of the sample. ThePT3100 has a powerful drive for higherrotational speeds that reduce processing time.Best of all the homogeniser can be cleanedquickly and simply under the tap without thescrubbing and scraping out required by othermodels.

Carbolite furnaces are used in research projects throughout the world wherever accurate, reliableheating up to 1800°C is required. Specialist models including three zone, vertical split, horizontalsplit, vacuum, rotating and rotary calciner are also available.Please ask for details from [email protected] or your local VWR sales organisation.

The Carbolite MTF tube furnace, which has a maximum temperature of 1200°C, is part of a specialrig used to assess the capacity, kinetics, thermodynamics and stability of different materials.Samples are tested in a sealed reactor that is heated at temperatures up to 500°C. The RisøNational Laboratory chose this furnace because of its compact design and consistently accurate,reproducible performance. The furnace has a 250 mm long heating zone with a diameter of 25 mm, and temperature is very accurately controlled through a PID instrument.

Fast chromatography

The LaChrom Elite® system can be operated in a wide flow range from micro HPLC with 1 mmi.d. columns up to 10 ml/min. For fastchromatography we recommend the use ofChromolith® columns that in contrast to particulate columns consist of one monolithicstationary phase, that generates a considerablylower back-pressure and faster elution atsimilar resolution. Chromolith® thereforepermits elevated flow rates (up to 9 ml/min)and a corresponding reduction in analysistimes, including when using flow gradients forlate eluting compounds. Even though higherflow rates are used, solvent consumption peranalysis is reduced saving consumable costs.

If higher resolution is needed it is possible tocombine several of the Chromolith® columnsdue to the low back-pressure they execute.Therefore, LaChrom Elite® in combination withChromolith® columns is the well-proven betteralternative to UPLC® that usually requires thepurchase of a special high-cost system for theextreme back-pressures created by thecolumns filled with 1,7 m particles! Moreover,high column temperatures for reduction ofsolvent viscosity are also not needed.

Fast automated HPLCmethod developmentLaChrom Elite® combined with the powerfulChromSword® Auto Software performscompletely automatic unattended HPLCmethod development and optimisation withinhours. Even existing methods can be optimisedand analysis time further reduced.

After some initial inputs ChromSword® Autopredicts method conditions and controls theLaChrom Elite® system to acquire the necessarychromatograms that are then analysed byChromSword® Auto to find the final optimum

conditions. The rapid optimisation mode givesan overview of the chromatographicperformance of the column/solvent systemwhile the detailed optimisation mode acquiresand analyses more data to predict and executeseveral alternative final optimum conditions.

For professional HPLC method developmentboth modes can be applied in combination with column and solving switching proceduresin order to try a variety of column/solventcombinations completely automatically.This is applied in method developmentlaboratories of pharmaceutical and chemicalcompanies especially using the rapidoptimisation mode for rapid screening ofcolumns and solvents, with the goal to quicklyfind a suitable separation system. The bestcandidate is then used for detailed methodoptimisation.

AutoRobust software is able to carry outrobustness studies for the elaborated optimummethods or existing methods completelyautomatically controlling the LaChrom Elite®

system and acquiring the chromatograms withthe desired small variations to the HPLCconditions.

These features make ChromSword® Auto andAutoRobust the acknowledged bestprofessional HPLC method developmentsoftware in the world.

Fast method validation

The LaChrom Elite® system and its softwareassistants also reduce the time and effortneeded for method validation via ValidationManager software that helps to create thevalidation plan, injection and samplepreparation tables, import chromatogram dataand calculate/evaluate method characteristics,enabling the production of a validation reportat the push of a button.

How will the success be quantified?What kind of testing is undertaken?

Such examinations of water quality of lakes and rivers depend on the requirements of legal

regulations but all starts with an on-sitesampling in the field.

First of all the most important physical andchemical data are measured using portableinstruments: pH, conductivity, dissolvedoxygen (saturation and/or concentration) and

temperature. For this purpose handhelds orportables such as ProfiLine 197i from

WTW are ideal. Multi parameterinstruments like Multi 350i withGLP supporting features and theappropriate sensors (combinedconductivity/DO probe ConOx or

pH/conductivity/DO probe MPP350) make these measurements

faster and more convenient withoutreducing the quality of the results.

A special measurement is the recording ofdepth profiles in lakes or at barrages. Depth

probes like TA 197 or MPP 350 allow a directmeasurement in comparison to a scoopedsample. Cable lengths up to 100 m cover a widerange of applications. Strong temperaturegradients that occur during summertime can bemonitored in situ as well as depth dependingoxygen concentrations. A sample of a pH, DOand temperature profile is given in the picturebelow.

Results on the one hand reflect the current situation and offer the chance to take measuresfor the future in the case of occurring problems;on the other hand they document thedevelopment and improvement of water quality.

Summary: besides sampling and laboratoryanalysis the on-site monitoring of water qualityis a very important factor. A direct measurementof depth profiles gives more accurate results incomparison to sampling; of course this is stillnecessary for an exact determination of chemicalparameters. Modern handhelds and portablesprovide results of highest accuracy andresolution. Multi parameter systems reduceefforts, handling and help to save time andcosts. Data logger and data transfer via PCinterfaces reduce errors produced by the“human factor”, especially when collecting dataoutside.

• Grow samples on GVPC/BCYE agarplates

• Pick 1 suspect Legionella colonyusing a loop or needle

• Suspend bacteria in 144 µl 0,9% NaCl(1% Tween) and add 16 µl Polymyxin

• Incubate 2-5 min in room temp

• Heat kill for 5 min at 95°C

• Cool and pipette 150 µl into the sample port of the Duopath
