Sorbent Selection Guide
AccuCATBond Elut AccuCAT cartridges are mixed-mode SPE columns consisting of a strong cation exchange and a strong anion exchange sorbent packed into one bed. AccuCAT is effective for the extraction of acidic, basic, and neutral compounds from urine and other biological samples. AccuCAT is particularly effective for catecholamine extraction from biofluids.
Alumina-A (AL-A)• Acidic Alumina (Al2O3) • pH: 4.5• Particle size: 25 µm• Primary retention mechanism: Lewis acid/base, polar, and ion exchange
Alumina is a classic Lewis acid, lacking two electrons in the aluminum center. The Lewis acid properties are enhanced in Alumina-A, making the sorbent more retentive towards electron-rich compounds. This sorbent has a slightly cationic nature through pretreatment with acidic solutions. The result is a surface that favors neutral and anionic species (e.g. neutral acid or acid anion), while discouraging retention of cationic species. Weak ion exchange is also possible with anionic compounds.
Alumina-B (AL-B)• Basic Alumina (Al2O3)• pH: 10.0• Particle size: 25 µm• Primary retention mechanism: Lewis acid/base, polar, and ion-exchange
Alumina-B has a surface that favors retention of cationic or hydrogen-bonding species. This sorbent has an anionic nature through the pretreatment of the alumina with basic solutions. The result is a surface that favors cation exchange, though the surface also exhibits some Lewis base properties and therefore can also retain electron-donors like neutral amines. However, this capacity is much lower than for the neutral and acidic aluminas. Strong hydrogen bonding is possible on Alumina-B, making it particularly effective for polar cations.

Alumina, like silica, is an extremely polar sorbent. The alumina surface tends to be slightly more stable under high pH conditions than unfunctionalized silica. The smaller particle size ensures high extraction efficiency and small (50 mg) sorbent beds are therefore commonly used. This sorbent has an electrically neutral surface that favors the retention of electron-rich compounds like aromatic species and aliphatic amines. It also favors retention of compounds containing electronegative groups (for example, functional groups with oxygen, phosphorus and sulfur atoms). It may be used in extracting either nonpolar or polar species from aqueous or nonaqueous samples respectively.
Aminopropyl (NH2)• Ideal for structural isomer fractionation• Anion exchange and polar retention mechanisms• Weak nonpolar retention
Aminopropyl (NH2), a polar sorbent, utilizes both hydrogen bonding and anion exchange. With a pKa of 9.8, NH2 is a weaker anion exchanger than sorbents such as SAX (a quaternary amine sorbent that is always charged) and is therefore a better sorbent choice for retention of very strong anions, such as sulfonic acids, which may retain irreversibly on SAX. Because a propyl tether supports the NH2 functionality, it can be used for nonpolar isolations from polar samples, but its strong polarity is its primary characteristic. NH2 is excellent for separation of structural isomers.
AtrazineBond Elut Atrazine is specially designed for the extraction of Atrazine and its desalkylated metabolites -- des-ethylatrazine and desisopropylatrazine -- from water samples. The sorbent is based on Bondesil C18-LO, with carefully controlled carbon content to ensure reproducibly high recoveries of Atrazine and its polar metabolites. The bigger particle size of the sorbent and the diameter of the tube allow fast flow for large volume water samples. And, the large bed mass of the Bond Elut Atrazine phase has been optimized to provide enough capacity to ensure good recovery of Atrazine and its polar metabolites. With the use of the Bond Elut Atrazine extraction cartridge and optimized method (M2638) in combination with a Varian HPLC diode array detection system, quantitative recoveries of Atrazine and its desalkylated metabolites in surface water have been achieved, even at extremely low concentrations (0.02 ppb). These concentration levels are far below the tolerance level specified by the EPA (3 ppb) and by those set by the most stringent European agencies (0.1 mg/L and 0.5 mg/L). The extraction and analysis methods are fast, reproducible, and require minimal organic solvent consumption.
BioBond ElutChoose BioBond Elut for fast, simple desalting of synthetic oligonucleotides from final deprotection reaction mixtures. With BioBond Elut there are no minimum molecular weight limits, so oligos less than 5,000 Daltons can be desalted with >98% recoveries. And, while typical desalting media can leave as much as 2% of the original salt in your sample, BioBond Elut provides 100% salt-free samples.

Due to the single carbon functional group, Bond Elut C1 is the least retentive of all alkyl-bonded phases for nonpolar compounds. However, because the sorbent is endcapped, silanol activity is generally masked. The result is easier retention and elution of polar and multifunctional compounds.
C2 • Bonded functional group: ethyl• Primary retention mechanism: weakly nonpolar• Typical sample types: plasma, urine, and aqueous samples
Bond Elut C2 is generally considered more polar than longer-chain alkyl-bonded sorbents. The short C2 alkyl group provides a smaller area for nonpolar interactions to occur. C2 is often used as a replacement for C18 and C8 when molecules are retained too strongly by the latter. C2’s polarity is slightly lower than CN for polar interactions. It is popular for the extraction of drugs from plasma and serum because of the very flat chromatographic baselines that can be achieved with appropriate wash and elution conditions.SPEC C2 is a monolithic disk featuring an unendcapped dimethyl bonded functional group. SPEC C2, therefore, offers a different selectivity than Bond Elut C2.
C8• Bonded functional group: octyl• Primary retention mechanism: moderately nonpolar• Typical sample types: water, aqueous biological samples
Bond Elut C8 is very similar in properties to C18, but is not as retentive for nonpolar compounds due to its shorter hydrocarbon chain. C8 is a good replacement for C18 when analytes are too strongly retained on C18 for effective elution. The potential of C8 for polar interactions with analytes is somewhat higher than for C18 because the shorter hydrocarbon chain does not mask the silica surface as effectively, but polar interactions are still not a significant property of C8. The C8 sorbent has been successfully utilized in the simultaneous extraction of fat- and water-soluble vitamins from human serum.
SPEC C8 offers similar selectivity to Bond Elut C8.
C18• Bonded functional group: trifunctional octadecyl• Primary retention mechanism: strongly nonpolar• Typical sample types: water, aqueous biological fluids
Bond Elut C18 is the most hydrophobic silica-based sorbent available. It is the most popular SPE sorbent because of its extreme retentive nature for non-polar compounds. C18 is generally regarded as the least selective silica based sorbent, because it retains most organic analytes from aqueous matrices — often a benefit when the analytes vary widely in structure. The potential for polar interactions with analytes is less significant with C18 than with any other sorbent because of the predominant effect of the long hydrocarbon chain. When extracting small- to intermediate-sized molecules, C18 can also be utilized for desalting prior to ion exchange because salts pass through it unretained.SPEC C18 is a nonendcapped, monofunctional version of the octadecyl bonded phases and offers similar selectivity to Bond Elut C18-OH.
C18 AR• Bonded functional group: trifunctional octadecyl• Primary retention mechanism: strongly nonpolar• Typical sample types: water, aqueous biological fluids
SPEC C18AR (acid resistant) is an endcapped, trifunctional version of the octadecyl phases and offers similar selectivity to Bond Elut C18. C18AR is exclusively available in SPEC disc format.
C18 EWP• Bonded functional group: trifunctional octadecyl• Primary retention mechanism: strongly nonpolar• Typical sample types: water, aqueous biological fluids
Bond Elut C18 EWP (extra wide pore) sorbent has 500 Å pores to allow more efficient extraction of large molecules (> 15,000 MW). Macromolecules of this size are typically excluded from standard porosity phases, but with C18 EWP successful separation of complex mixtures of proteins, peptides or nucleotides is achieved. C18 EWP is a good sorbent for desalting large molecular weight compounds prior to LC/MS or LC/MS/MS, capillary electrophoresis, or any other analytical technique that is adversely affected by the presence of salts in the sample.

Bond Elut C18 INT (intermediate carbon loading) is similar in properties to the standard Bond Elut C18 sorbent, but has a slightly lower carbon loading. This intermediate carbon load C18 is thoroughly endcapped, ensuring rapid release of basic drugs. The carbon loading has been optimized to differentiate between metabolites of classes of compounds such as prostaglandins. In addition, the narrow pore size helps exclude large molecules such as proteins.
C18 LO• Low carbon loading gives unique C18 selectivity• Strongly nonpolar retention mechanism• Ideal for polar analyte extraction from water samples and
biological fluids
C18 LO, like C18 and C18 INT, is a highly retentive nonpolar silica-based sorbent due to the long hydrocarbon chain. However, the low carbon loading of C18 LO gives unique selectivities over other octadecyl sorbents. The carbon loading has been optimized for polar analyte extraction from environmental matrices. Quantitative recoveries of Atrazine metabolites have been successfully achieved with C18 LO sorbents. (Note: see Bond Elut Atrazine section for optimized bed mass and application information.)
C18 OH• Bonded functional group: monofunctional octadecyl• Primary retention mechanism: moderately nonpolar, plus significant
polar secondary interactions• Typical sample types: biological matrices, aqueous samples
Bond Elut C18 OH is a non-endcapped version of the octadecyl bonded phases that enable the silanols on the silica surface to be more active. This low-load C18 has controlled silanol activity under tight QC specifications for superior batch-to-batch reproducibility. The silanol activity permits fractionation of metabolites and enhances retention of basic compounds compared with endcapped C18. In addition, the 150Å intermediate pore size is ideal for intermediate molecular weight compounds that might not be retained on a standard 60Å material.SPEC C18 offers similar selectivity to Bond Elut C18 OH.
Carboxylic Acid (CBA)• Bonded functional group: carboxylic acid• Primary retention mechanism: weak cation exchange• Best choice for extraction of strong cations• Typical sample types: water, biological fluids
Bond Elut CBA is a medium polarity sorbent based on carboxy-propyl-functionalized silica, and exhibits either polar or nonpolar interactions depending on the matrix or solvent environment. Its primary characteristic is its weak cation exchange ability. Because of its pKa of 4.8, CBA does not require the high basic conditions for elution that are required by strong cation exchange sorbents. Simply, lowering the pH to 2.8 effectively neutralizes CBA, permitting elution of the retained analytes. For this reason, CBA is often the best choice for extraction of very strong cations.

Bond Elut Certify is the original mixed-mode sorbent, designed for the extraction of basic (cationic) drugs from urine, plasma, serum, whole blood and other biological matrices. Examples of drugs and drug classes that can be extracted with Certify include:• Basic and nonpolar pharmaceutical compounds• HC metabolite, 11-nor-delta-9-tetrahyrdocannabinol-9- carboxylic acid• Cocaine and benzoylecognine• Amphetamine and methamphetamine• Phencyclidine• Opiate and 6-MAM• Barbiturates• Benzodiazepines• Acid/base/neutral screens
Analytes are strongly retained on Certify by a combination of ion exchange and nonpolar mechanisms. This allows the use of aggressive aqueous and organic washes to rinse away nonpolar, polar and ionic matrix contami-nants. The result is outstanding sample clean up and concentration of samples prior to screening or confirmation by GC, GC/MS, LC/MS or TLC.
SPEC MP-1 offers similar selectivity to Bond Elut Certify.

Bond Elut Certify II was developed specifically for the rapid and effective extraction of acidic drugs and metabolites from urine and other biological matrices. Certify II is a mixed-mode cartridge packed with nonpolar C8 and strong anion exchange (SAX) sorbent. It has been optimized for extraction of acidic and nonpolar drugs such as THC metabolite and nonsteroidal anti-inflammatory drugs.
Cyclohexyl (CH)• Bonded functional group: cyclohexyl• Primary retention mechanism: moderately nonpolar• Typical sample types: water, aqueous samples
Bond Elut CH is a medium polarity sorbent that exhibits unique selectivity in nonpolar extractions. When employed as a nonpolar sorbent, CH has the approximate polarity of a C2 sorbent. The polar subsurface is an important factor among CH properties, as indicated by the ability of CH to retain polar analytes such as phenol from aqueous matrices. Because of its unique selectivity, CH is often a good choice when other nonpolar sorbents do not provide the desired selectivity.
Cyanopropyl (CN-E)• Bonded functional group: endcapped cyanopropyl• Primary retention mechanism: moderately nonpolar (aqueous matrix)
or polar (nonpolar organic matrix)• Typical sample types: aqueous (nonpolar), organic (polar)
Bond Elut CN-E is a medium polarity sorbent ideal for extraction of extremely nonpolar compounds that irreversibly retain on more nonpolar sorbents such as C18 or C8. The endcapped version of the cyano sorbent is best utilized when extracting analytes from an aqueous matrix.

Unendcapped Bond Elut CN-U is a medium polarity sorbent. It is less retentive than Si or 2OH for very polar analytes that might be retained irreversibly on these more polar sorbents. The unendcapped cyano phase is ideally suited for the extraction of polar compounds from a nonpolar matrix such as hexane or oils.
SPEC CN offers similar selectivity to Bond Elut CN-U.

Bond Elut DEA bears some resemblance to PSA and NH2 in its properties. It has slightly lower capacity as an anion exchange sorbent (1.0 meq/g), and a more nonpolar character due to a longer alkyl tether. DEA is somewhat more polar than C8, but less polar than C2 or CN.
Diol (2OH)

Diol (20H) is typically used for polar extractions from nonpolar solvents. Diol resembles unbonded silica in its tendency for strong hydrogen bonding. It also shares silica’s important ability to discriminate between similar compounds such as structural isomers. Prostaglandins, for example, can be separated into classes using Diol by slightly varying the solvent polarity. Diol is a more predictable sorbent than silica for extraction methods requiring a polar sorbent conditioned with a polar solvent. Diol can also be used for nonpolar retention by virtue of its hydrocarbon spacer.

Florisil is a magnesia-loaded silica gel. Like silica, it is extremely polar in nature and ideal for the isolation of polar compounds from nonpolar matrices. The larger particle size – 200 µm – of the Florisil material enables fast flow of large-volume samples and thus is a potential alternative to silica when extracting viscous samples. Florisil is ideal for the separation of chlorinated pesticides, and is used in numerous AOAC- and EPA- regulated methods. Florisil has also been used as a replacement for Alumina in cases where the Lewis-acidity of alumina interferes with the extraction.

Phenylboronic Acid (PBA)• Strong covalent retention mechanism• Multiple secondary interactions• Ideal for cis-diol extraction
PBA (Phenylboronic Acid) is a unique sorbent consisting of a phenylboronic acid bonded to a silica gel surface. The PBA sorbent covalently binds cis-diol, cis-amino alcohols, and other related functional groups. Because covalent retention involves an interaction of 10-100 times greater energy than other extraction mechanisms, compounds are more tightly bound to the sorbent surface, resulting in superior clean-up. The boronate group has a high specificity for cis-diol containing compounds such as catechols, nucleic acids, low molecular weight proteins and carbohydrates. PBA is especially effective in the isolation of catecholamines from biological fluids and for the separation of RNA from DNA. It may also be applied to cis-amino alco-hols such as propranolol. Retention is usually strongest when the analytes’ functional groups are coplanar.
Phenyl (PH)• Bonded functional group: phenyl• Primary retention mechanism: moderately nonpolar• Typical sample types: water, biological fluids
Bond Elut PH is most commonly employed for nonpolar extractions, with a similar polarity to a C8 sorbent. Like cyclohexyl, PH exhibits a slightly different selectivity from other nonpolar sorbents. This added selectivity results from the electron density of the aromatic ring. Retention of planar, conjugated organic molecules is enhanced compared to the aliphatic bonded phases.
SPEC PH offers similar selectivity to Bond Elut PH.

Bond Elut PRS is a strong cation exchange sorbent that is also very polar. As such, PRS exhibits weak nonpolar interactions. The pKa of PRS is low, and cationic analytes must be eluted either by high ionic strength, or by neutralizing the charge on the cationic analyte. Therefore, PRS is most effective for weaker cations such as pyridinium compounds. Also, PRS is recommended where nonpolar secondary actions of SCX interfere with the extraction.

Bond Elut PSA is an anion exchange sorbent that is similar to NH2. PSA has two amine groups that offer slightly greater ionic capacity (1.4 meq/g) than NH2 (1.1 meq/g), and the pKas of its primary and secondary amine groups are higher (10.1 and 10.9, respectively). PSA is therefore a stronger anion exchanger than NH2. The PSA functional group is also a very good bidentate ligand, making PSA an excellent sorbent for chelation applications (useful for metal ion extractions). Its higher carbon loading makes it a less polar sorbent than NH2, and thus a better choice for very polar compounds that retain too strongly on NH2 sorbent.SPEC PSA offers similar selectivity to Bond Elut PSA.
SAX• Strongest anion exchanger available• Quaternary ammonium chloride for intermediate selectivity
SAX (Strong Anion eXchange) is the strongest anion exchange sorbent available. Because its functional group is a quaternary amine, the sorbent is always charged. SAX offers minimal nonpolar interactions because any effect of the carbon atoms in the functional group is masked by the amine. Because SAX is such a strong anion exchanger, it is a good sorbent for retention of weaker anions such as carboxylic acids that may not retain strongly enough on weaker anion exchange phases. SAX contains a chloride counterion. The user can modify the selectivity of SAX by conditioning with appropriate buffers, exchanging chloride for ions of greater or lesser selectivity.
SCX• Bonded functional group: benzenesulfonic acid• Primary retention mechanism: strong cation exchange• Typical sample types: aqueous, biological fluids and organic reaction

Bond Elut SCX is a strong cation exchanger with a very low pKa, and is similar to PRS as an ion exchange sorbent. The SCX sorbent has a higher potential for nonpolar interactions than PRS due to the presence of the benzene ring. This potential for nonpolar interactions should be considered when the sorbent is used for ion exchange from aqueous samples. This dual nature is useful for extracting compounds that exhibit both cationic and nonpolar character. The overall result of the dual character is superior clean up using only a single sorbent.SPEC SCX offers similar selectivity to Bond Elut SCX.
Silica (SI)• Sorbent description: native silica, unfunctionalized• Primary retention mechanism: strongly polar• Typical sample types: nonpolar organic solvents, oils, and lipids
Silica is generally regarded as the most polar sorbent available. Unbonded, activated silica is also fairly acidic. SI is one of the best sorbents available for selectively separating compounds of very similar structure. Applying the analytes in a nonpolar solvent, then gradually increasing the solvent polarity in successive wash steps by adding increasing concentrations of polar modifiers, such as THF or ethyl acetate, can accomplish this separation.
Bond Elut SI and SPEC SI offer similar selectivity.
TCA• Optimized for tricyclic antidepressant (TCA) extraction• Broad concentration range (20-2000 ng/mL)
TCA is a nonpolar sorbent optimized for extraction of tricyclic antidepressants (TCAs). Each lot of Bond Elut TCA is specifically quality tested for recovery of TCAs, ensuring reproducibly good performance.
TOPTOP (Trityl-on Oligo Purification) was created with a very high selectivity for trityl-on oligonucleotides. It is based on a unique functionalized styrene divinylbenzene material that allows direct loading of the basic deprotection solution. This ensures that all the full-length oligonucleotide product will bind the first time it is passed through the TOP cartridge. FOCUS’ improved retention for polar analytes allows use of a strong Power Rinse without causing drug loss. Rigorous washes using 10-20% acetonitrile and methanol can be used to aggressively remove endogenous matrix interferences. For very polar analytes that are held by strictly polar interactions, e.g. atenolol, washes of up to 100% can be used. Empore® Disk ManifoldsThe one-, three- or six-station extraction vacuum manifold offers an efficient extraction apparatus. These manifolds allow multiple sample processing and are more stable and compact than a series of single glass filtration systems. Using the manifold, up to six extractions can be completed simultaneously, and a single operator can manage multiple manifolds. The powerful combination of the Empore disk and the six station manifold allows for up to 12 one-liter drinking water samples to be extracted per hour, a tremendous improvement over manual liquid/liquid methods.
The Empore Extraction Manifold accommodates a set of six 47 mm or three 90 mm glassware. Replacement parts may be ordered separately. A vacuum pump or supply is required.

Alumina, like silica, is an extremely polar sorbent. The alumina surface tends to be slightly more stable under high pH conditions than unfunctionalized silica. The smaller particle size ensures high extraction efficiency and small (50 mg) sorbent beds are therefore commonly used.
This sorbent is prepared with an electrically neutral surface. This favors the retention of electron-rich compounds like aromatic species and aliphatic amines. It also favors retention of compounds containing electronegative groups (for example, functional groups with oxygen, phosphorus and sulfur atoms). It maybe used in extracting either non-polar or polar species from aqueous or non-aqueous samples respectively.

Bond Elut Atrazine
Atrazine is a widely used herbicide in the United States that has been implicated in serious health problems, including tumor formation and endocrine problems. The biodegradation of atrazine and the harmful affects on the environment by some of its dealkylated metabolites, de-ethylatrazine and deisopropylatrazine, have placed these herbicide degradation products in the “watch list” of the National Pollutants Survey (NPA).
The Varian Bond Elut Atrazine cartridge is a specially bonded low-load, high-flow C18 phase. The controlled carbon content of the sorbent was designed to retain atrazine as well as its polar metabolites by a non-polar/polar retention mechanism. The bigger particle size of the sorbent and the diameter of the tube allow fast flow for large volume water samples. And, the large bed mass of the Bond Elut Atrazine phase has been optimized to provide enough capacity to ensure good recovery of atrazine and its polar metabolites.
With the use of the Bond Elut Atrazine extraction cartridge and optimized method for surface water (M2638) in combination with a Varian HPLC diode array-detection system, quantitative recoveries of atrazine and its dealkylated metabolites have been achieved, even at extremely low concentrations of 0.02 ppb. These concentration levels are more than two orders of magnitude below the tolerance level specified by the EPA (3 ppb) and those set by the most stringent European agencies (0.1 mg/L and 0.5 mg/L). The extraction and analysis methods are fast, reproducible and require minimal organic solvent consumption.

Bond Elut C1 sorbent
Due to the single carbon functional group, C1 is the least retentive of all alkyl group bonded phases for nonpolar compounds. However, because the sorbent is endcapped, masking the polar silanol activity, easier retention and elution of polar and multifunctional compounds can be achieved.

Bond Elut C2 sorbent
C2 is a fairly polar sorbent because of the short chain length of the functional group, which exposes the silica surface. C2 is often used as a replacement for C18 and C8 when molecules are retained too strongly by the latter. C2’s polarity is slightly lower than a CN for polar interactions. It is popular for the extraction of drugs from plasma and serum since, if correct wash and elution conditions are selected, flat baselines can be achieved.

Bond Elut C8 sorbent
C8 is very similar in property to C18, but is not as retentive for nonpolar compounds due to its shorter hydrocarbon chain. C8 is a good replacement for C18 when analytes are too strongly retained on C18 for effective elution. The potential of C8 for polar interactions with analytes is somewhat higher than for C18 because the shorter hydrocarbon chain does not mask the silica surface as effectively, but polar interactions are still not a significant property of C8. The C8 sorbent has been successfully utilized in the simultaneous extraction of fat- and water-soluble vitamins from human serum.

Bond Elut C18 sorbent
C18 is the most hydrophobic silica-based sorbent available. It is the most popular SPE sorbent because of its extreme retentive nature for nonpolar compounds. C18 is generally regarded as the least selective silica based sorbent, since it retains most organic analytes from aqueous matrices, often a benefit when the compounds of interest vary widely in structure. The potential for polar interactions between analytes and sorbent is less significant with C18 than with any other sorbent because of the predominant effect of the long hydrocarbon chain. When analyzing small to intermediate molecules, C18 can also be utilized for desalting aqueous matrices prior to ion exchange because salts pass through it unretained.

Bond Elut C18 sorbent
The C18 EWP sorbent is based upon standard particle size silica but with 500 Å pores to allow more efficient extraction of macromolecules typical of most bimolecular separations. The larger pore size of the sorbent enables large molecules (> 15,000 MW), which are typically excluded from standard porosity phases, to permeate into the pores and interact with the bonded silica surface. With C18 EWP, successful separation of complex mixtures of proteins, peptides or nucleotides can be achieved. Also, C18 EWP is a good sorbent choice for desalting matrices with large molecular weight com-pounds of interest prior to LC/MS or LC/MS/MS, capillary electrophoresis or any other analytical technique that is adversely affected by the presence of salts in the sample. Bond Elut C18 sorbent
C18 INT is very similar in properties to the standard Varian C18 sorbent but with a slightly lower carbon loading. This intermediate load bonded C18 is also thoroughly end capped ensuring rapid release of basic drugs. The carbon content has been optimized to differentiate between metabolites of species like prostaglandins. In addition, the narrow pore size helps exclude large molecules such as proteins.

C18 LO, like C18 and C18 INT, is a highly retentive nonpolar silica-based sorbent due to the long hydrocarbon chain. However, the low carbon loading of C18 LO gives unique selectivities over other octadecyl sorbents. The percent carbon loading has been optimized for polar analyte extraction from environmental matrices. Bond Elut C18 sorbent
C18 OH is a nonendcapped version of the octadecyl bonded phases that enables the silanols on the silica surface to be more active. This low-load C18 has controlled silanol activity under tight QC tolerances for best batch-to-batch reproducibility. The silanol activity permits fractionation of metabolites and enhances retention of basic compounds compared with endcapped C18. In addition, the 150 Å intermediate pore size is good for intermediate molecular weight compounds that may not be retained on a standard 60 Å material.

Bond Elut CBA carboxylic acid sorbent
CBA is a medium polarity sorbent based on carboxy propyl functionalized silica, exhibiting either polar or nonpolar interactions depending on the matrix or solvent environment. Its prime characteristic is its weak cation exchange properties. Because of its pKa value of 4.8, CBA does not require extreme basic conditions for elution that are required by other cation exchange sorbents. Above a pH of 4.8, the majority of functional groups carry a negative charge that can be used for retaining cationic compounds. Dropping the pH to 2.8 effectively neutralizes CBA, permitting the retained analytes to elute from the sorbent. For this reason, CBA is often the best choice for cation exchange, especially when dealing with very strong cations (i.e. high pKa cations). In addition, CBA shows a wider range of selectivity towards counter-ions than other cation exchange phases, enabling more options for elution.

Bond Elut® Cellulose
The Varian Bond Elut Cellulose column utilizes a pure microgranular cellulose powder that is packed between two 20 µm polyethylene frits in an ultraclean 3 mL polypropylene extraction column. The unique rod-like shape and controlled particle size of this variety of Cellulose ensure that it is free-flowing when dry. In addition, the material is manufactured from high purity cotton with a very high a-cellulose content that ensures optimum column packing properties. Thus, high density, more uniform, smooth-packed column beds are achieved. In addition, the cellulose phase is very stable over a wide pH range and contains an extremely low metal content (Fe and Cu content less than 5 ppm). The combination of surface area and polymeric structure results in a sorbent with excellent capacity.
The cellulose media contains numerous hydroxyl groups. Because of its polar nature, it is able to accept high loading of many polar substances from aqueous and organic phases. The free hydroxyl groups’ binding properties are complemented by the presence of very small quantities of residual aldehydes, ketones and carboxyl groups. These weak groups add reducing and ion-exchange properties.
This microgranular cellulose support has been utilized for large scale bioprocessing as well as a prefilter for suspended matter (i.e. cell debris and suspended fats), as a prefractionation step prior to reverse phase HPLC, for separation of biomolecules, and as an affinity chromatography matrix.

Bond Elut CH cyclohexyl sorbent
Cyclohexyl is a medium polarity sorbent that exhibits unique selectivities for certain analytes. When employed as a nonpolar sorbent, CH has the approximate polarity of a C2 sorbent. The polar subsurface is an important factor among CH properties, as indicated by the ability of CH to retain polar analytes such as phenol from aqueous matrices. Because of its unique selectivity, CH is often a good choice when other nonpolar sorbents such as C18, C8, C2, and PH do not provide the desired selectivity.

Bond Elut DEA diethylaminopropyl sorbent
Bond Elut DEA bears some resemblance to Bond Elut NH2 in its properties, as does Bond Elut PSA. DEA has slightly lower capacity as an anion exchange sorbent (1.0 meq/g) than NH2, and a more nonpolar character due to the additional carbon chain on the functional group. NH2, a very polar sorbent, can utilize both hydrogen bonding and anion exchange. Since the pKa of the NH2 sorbent is 9.8, at any pH below 9.8, the majority of the functional groups are positively charged. NH2 is a weaker anion exchanger than sorbents like SAX (a quaternary amine sorbent that is always charged) and is therefore a better sorbent choice for retention of very strong anions, such as sulfonic acids, which may be retained irrevers-ibly on SAX. Because an ethyl group supports the NH2 functionality, it can be used for nonpolar isolations from polar samples, but its strong polarity is its primary characteristic. Like 2OH and SI, NH2 is excellent for separa-tion of structural isomers.

PBA is a unique sorbent consisting of a phenylboronic acid covalently linked to a silica gel surface. Because covalent retention involves an interaction with 10-100 times greater energy than other extraction mechanisms, compounds are more tightly bound to the sorbent surface resulting in a more complete cleanup. The boronate group has a high specificity for cis-diol-containing compounds like catechols, nucleic acids, low molecular weight proteins and carbohydrates. PBA has proven to be especially effective in the isolation of catecholamines from biological
fluids, and for the separation of RNA from DNA. Also, it may be applied to cis-amino alcohols such as propanol. Retention is usually strongest when the analytes’ functional groups are coplanar.

A strong cation exchange sorbent that is also very polar, PRS does not exhibit any appreciable degree of nonpolar interactions. In nonpolar solvents, PRS is capable of polar and hydrogen bonding interactions. As the pKa of PRS is low, cationic compounds must usually be eluted either by a high ionic strength eluant, or by neutralizing the charge on the cationic isolate. Therefore, PRS is most effective for weaker cations such as pyridinium compounds. The rela-tive lack of nonpolar secondary interactions, combined with its low capacity, gives PRS a unique set of selectivity properties.

Bond Elut PSA ethylenediamine-N-propyl sorbent
PSA is similar to NH2. An anion exchange sorbent, PSA has two amine groups that offer higher ionic capacity than NH2 (1.4 meq/g vs. NH2’s 1.1 meq/g), and the pKas of its primary and secondary amine groups are higher (10.1 and 10.9, respectively). PSA is therefore a stronger anion exchanger than NH2. The PSA functional group is a very good bidentate ligand, making PSA an excellent sorbent for chelation. Its higher carbon content makes it a more nonpolar sorbent than NH2 and thus a better choice for very polar compounds that retain too strongly on NH2 sorbent.

Bond Elut SAX trimethylaminopropyl sorbent
SAX is the strongest anion exchange sorbent available. Because its functional group is a quaternary amine, the sorbent is always charged. SAX offers minimal nonpolar interactions because any effect of the carbon atoms in the functional group is masked by the amine. Because SAX is such a strong anion exchanger, it is a good sorbent choice for retention of weaker anions such as carboxylic acids that may not retain strongly enough on weaker anion exchange phases. It is supplied in the chloride counter-ion form. The user may modify its selectivity by conditioning it with appropriate buffers, as described in The Handbook of Sorbent Extraction Technology (P/N 12245005).

SCX is a strong cation exchanger with a very low pKa, similar to PRS. However the SCX sorbent has a higher potential for nonpolar interactions than PRS, due to the presence of the benzene ring on its surface. This nonpolar char-acter of SCX should be considered when the sorbent is used for ion exchange from aqueous solvent systems. This dual nature is useful with compounds that exhibit both cationic and nonpolar character. The overall result of the dual character is superior clean up using only a single sorbent.

Bond Elut SI silica sorbent
Silica is generally regarded as the most polar sorbent available. Unbonded, activated silica is also fairly acidic. SI is one of the best sorbents available for selectively separating compounds of very similar structure. Applying the analytes in a nonpolar solvent, then increasing the solvent polarity by adding increasing concentrations of polar modifiers, such as THF or ethyl-acetate, at successive steps can accomplish this separation.

Polymer and C18 Varieties Application Development KitIn response to the popularity of high-throughput screening, Varian of-fers a polymer and C18 method development kit that contains our wide range of C18 silicas and nonpolar resins. In addition, both the standard (40 µm) particle size and a high flow (120 µm) C18 phase have been included. This kit will allow labs seeking to validate screening methods on nonpolar sorbents to find the best sorbent option. Each phase is packed into 1 mL straight barrel Bond Elut cartridges, which are compatible with robotic systems. Standard 20 µm, polyethylene frits are used. The 100 mg bed masses are optimized for smaller samples and elution volumes. The sorbents included in this kit are:
C18 - Our most widely used nonpolar sorbent: New applications are continually being developed on Bond Elut C18.
C18 HF -This 120 µm high flow sorbent is effective for viscous or particulate-laden samples, but also offers the added selectivity of silanols on the silica substrate.
C18 EWP - Extra wide pore size (500 Å) allows for better retention of macromolecules, proteins, and other large molecules typically excluded from standard 60 Å pores.
C18 OH - Monofunctional bonding provides slightly lower carbon loading, enhanced polar characteristics, and greater access to silanols, resulting in a unique selectivity for nonpolar analytes.
PPL - A functionalized polystyrene-divinylbenzene polymer (PSDVB), PPL is our most nonpolar sorbent. It is effective in extracting the most polar organic compounds from aqueous matrices.
ENV - The larger particle size of this pure PSDVB resin makes ENV useful for particulate-laden samples. In addition, it is useful for applica-tions requiring extremes of pH.

When a sample volume exceeds the capacity of a Bond Elut cartridge reservoir, you can increase the volume capacity by attaching a larger reservoir to the top of the smaller cartridge using a Bond Elut adapter. Bond Elut adapters fit on top of any Bond Elut cartridge and contain a female Luer fitting that accommodates the tip of another cartridge. This allows you to:
• Stack cartridges for custom multi-sorbent methods (#1 in the photo). • Increase any cartridge’s volume capacity by stacking an empty reservoir
on top of the cartridge (#2, 3). • Use a standard Luer-tipped syringe to apply positive pressure for
processing samples (#4). Luer-tipped syringes fit into any Bond Elut adapter. Gentle pressure is all that is needed to apply conditioning solvents, samples, rinse solutions, and elution solvents.
• Pull large-volume samples through any SPE cartridge (#5). Simply insert one end of a 1/8” OD tube through the opening in the Bond Elut adapter (forming a seal), and insert the other end of the tube into the sample container. Then apply a vacuum to the SPE cartridge to pull the entire sample through.
To select the appropriate adapter, choose the adapter specified for the size of the lower Bond Elut cartridge. The adapter fits snugly into the top of the cartridge reservoir. All Bond Elut cartridges and empty SPE cartridges have a standard Luer tip that is usable with these adapters.

Varian’s EnvirElut1664 Oil & Grease solid phase extraction processing system is ideal for monitoring waste water, aqueous effluent, and surface/ground water. It is faster, more efficient, and uses less solvent than sample preparation performed with liquid/liquid extraction. EnvirElut 1664 cartridges consistently pass 1 liter dirty water samples in less than 10 minutes without loss of analyte, delivering greater than 90% recovery with oil and grease standards from 5 mg/L to 1 g/L. Our cartridge design has been optimized for maximum flow of large-volume samples, and the use of depth filters ensures that even particulate-laden samples are processed without clogging. The procedure is entirely Freon-free, and offers a solid phase extraction alternative with results equivalent to EPA 1664/ASTM Method 413.1. A wide variety of disposable syringe-tip filters are available for applications requiring the highest quality membrane-based devices. Use the following selection guide for selecting the appropriate syringe filter materials:
Hydrophilic Nylon — naturally hydrophilic, high protein binding, high mechanical stability, and high dirt loading capacity. Often used for samples directly injected into HPLC sample loops.
Polypropylene (PP) — hydrophilic, low protein binding, and designed for aqueous and organic solvent based samples. This filter has low extractables and provides exceptionally low detection levels.
Regenerated Cellulose (RC) — hydrophilic, solvent-resistant membrane. It is a very low protein binder and is ideal for filtration of proteins, peptides and other biomolecules.
PVDF (polyvinylidene difluoride) — hydrophilic membrane suitable for most general-purpose filtration applications. It is a low protein binder and has a broad chemical compatibility.
Hydrophobic PTFE (Teflon®) — hydrophobic, low protein binding, recommended for HPLC solvent or gas chromatography filtration, aggressive solutions, and small venting applications. Requires prewetting for aqueous samples

Combilute Combilute is a 96-well SPE plate filled with Hydromatrix-brand diatomaceous earth. Each well is filled with approximately 250 mg Hydromatrix and sealed with a piercable sealing mat.
The product was specifically designed for drug discovery and pharmacokinetics laboratories performing:• Purification of combinatorial libraries• Screening for pharmaceuticals from biological fluids
Combinatorial Chemistry:Combilute is an ideal purification tool for combinatorial libraries. Remove polar reaction byproducts, reagents, and impurities. No vacuum manifold is required and the use of any standard robotic liquid handler can be used.
Pharmaceutical Drug Screening:Drug screening of plasma or other biological fluid samples can be efficiently performed with Combilute. In studies where a large number of patient samples must be quickly analyzed, the 96-well format of Combilute is indispensible. Robotic liquid handling systems, already used for many other functions in pharmaceutical labs, can be easily adapted to dispense 96 samples quickly into a single Combilute plate, positioned on top of a deep well collection plate. No vacuum manifold is required. And the entire procedure, from sample application to elution, can be easily and fully automated.

Chem Elut cartridges are ideal for purification or sample preparation.
For sample preparation of biological samples, select the cartridge which fits the total volume of the sample (including any buffers or additives). Simply load the sample; wait 3-5 minutes; then elute with organic solvent. This general method is ideal for LC/MS sample preparation.
For organic reaction mixture purification, simply load the cartridge with aqueous acid to scrub amines (or add base to scrub acids). Select cartridge size based on the total volume of aqueous buffer to be used. Then pass the crude synthesis mixture through the cartridge. Gravity flow is all that’s needed – no vacuum manifolds or pressure required. It’s the fast, easy, reproducible way to scrub organic synthesis mixtures of excess reagents and reaction byproducts!

Positive Pressure ManifoldPositive pressure is often preferable to vacuum for SPE sample processing. This is because:• Positive pressure provides more than two times greater total force for
moving viscous samples than vacuum• Positive pressure will not dry out SPE cartridges when viscous or other
difficult samples are being processed• Use of nitrogen or other inert gas for positive pressure applications
can improve recovery and extraction reproducibility or easily -oxidized analytes
The Varian Positive Pressure Mani-fold ensures fast and reproducible parallel processing of up to 24 Bond Elut cartridges (1, 3, 6, or 10 mL sizes). Pressure at each cartridge (up to 50 psig) is automatically controlled with individual valves that shut off flow when the cartridge is drained. This prevents faster flowing cartridges from drying out, so there’s no need for any manual intervention, such as shutting off stopcocks, which is typically done with vacuum manifolds. The Varian

Engineered to increase laboratory productivity, the corrosion-resistant Vac Elut 20 allows simultaneous processing of up to 20 Bond Elut cartridges. The manifold’s clear glass base allows careful monitoring of the entire sample collection process. Its compact, linear design requires very little benchtop space.

Rugged constructionOn the Vac Elut 20, the vacuum-control valve, vacuum gauge, and quick-release valve are mounted on the lid, away from the corrosive solvent stream and within convenient reach. The solvent-resistant polypropylene rack is available in a variety of sizes to accommodate the collection tubes commonly used in sample preparation.
Freedom from sample contaminationTo minimize the risk of sample carryover, low-cost, disposable, medical-grade polypropylene delivery needles can be easily replaced. Polypropylene extender tips are also available as a replacement for the standard needle valves, ensuring a direct path into the collection tube. Vac Elut® SPS 24 ManifoldThe Vac Elut SPS 24 allows you to process up to 24 SPE cartridges simultaneously. Like all Vac Elut manifolds, the SPS 24 is made from durable, solvent-resistant materials and engineered to last. The glass sides allow easy viewing of the entire sample collection process.
The highlighting feature of the SPS 24 system is its waste diversion funnel that enables all steps of the SPE procedure to be completed without removing the lid. The collection rack can be placed inside the unit before the extraction begins. This unique characteristic of the SPS 24 eliminates splash-back, prevents cross contamination, and minimizes hazardous waste, and biohazard exposure.

SPME Fibers
SPME fiber assemblies can be reused for up to 100 analyses or more, depending on the application and the care they are given. For reuse, simply condition with solvent or heat before and after every analysis. Each assembly has a color-coded or notched hub indicating the type of coating on the fiber. Choose the assembly that is appropriate for the holder: manual, or Varian AutoSampler (also used for HPLC applications). First time SPME users must order both a holder and a fiber assembly.
