    4Measure the blank

Press the measurement key. The device will display please insert blank.

Take a new iCheck Chroma reagent vial. Clean the vial with a paper tissue before inserting.

Be sure to only hold the vial by its metal top.

Take the vial you used for blank measurement and proceed with sample injection.

Your iCheck Chroma will request during each mea-surement that you perform blank measurement. Always use a new vial for the blank measurement and then use same vial to inject your sample.

Remove the metal cap and insert the vial into the measurement chamber of the iCheck.

Press the measurement key again. Wait until the iCheck displays Prepare for measurement.

  • 85Prepare your sample

iCheck Chroma quantitatively analyzes vitamin A in refined edible oil fortified in the range of 3 - 30 mg retinol equivalents (RE)/kg (10 - 100 interna-tional units (IU)/g).

Mix your oil sample well. Use a new syringe with-out a needle attached and take up approx. 0.7 mL of your oil sample.

Clean the end of the syringe with a paper tissue. Place the needle on the syringe. Hold the syringe with needle pointing up and gently tap on the syringe with your fingers to get the air bubbles to move up.

Adjust the volume of the sample to exactly 0.5 mL (500 L) by ejecting excessive volume into the pa-per tissue. Make sure no air bubbles are left inside.

    6Inject your sample

Slowly inject the sample into the iCheck Chroma reagent vial used for blank measurement.

Take special care to inject the oil sample slowly along the side of the glass and above the clear liquid in the vial in order to produce two distinct layers.

If you have injected the sample correctly, the content of your vial should look very similar to the image below. The oil will form a thin layer on top of the clear reagents in the vial. Make sure not to shake the vial yet.

    9Result display

iCheck Chroma will now analyze your sample. This will take approx. 30 seconds.

Do not move the vial or the iCheck until the result is displayed.

Sample result The result is displayed in mg RE/kg. Multiply your

result by 3.33 to convert from mg RE/kg to IU/g.

    CHROMA

    Batch 1, Sample 18.55 mg RE/kg

If your sample has under 3 mg RE/kg the device will display: Out of linear range. You will observe no blue color in the reagent vial.

If your sample has over 30 mg RE/kg the device will display: Out of linear range. You will observe intense blue color in the reagent vial.

    CHROMA

Batch 1, Sample 1Out of Linear Range

    (Range: 3 - 30 mg RE/kg)

In case your sample has over 30 mg RE/kg dilute the sample with unfortified refined edible oil. Multiply the result by the dilution factor (DF).

    DF = (fortified oil, g + unfortified oil, g)/fortified oil, g.

Note: For conversion of mg RE to IU please refer to the following factors: 1 mg RE = 1 mg retinol = 3333 IU 1000 IU = 0.3 mg RE.For support with dilution and calculation please contact BioAnalyt Support.

    10Data storage

For documentation purposes, iCheck Chroma has an internal memory to store up to 100 individual measurements including such information as batch number, sample number, date, time, and result.

For detailed description of the data transfer to a computer please refer to the Data Transfer section.

    11Disposal

Reagent vials contain hazardous chemicals and are disposed of according to national regulations for hazardous material. Collect the vials in a con-tainer and hand them over to a chemical waste company. Material safety data sheet (MSDS) of the reagent vials is provided with each shipment.

Take extra care when disposing of the used needles to prevent injury: discard used needles into special container.

    TechnicalData

iCheck Chroma is a state-of-the-art portable photom-eter for measuring vitamin A concentration in edible oil. The iCheck is specifically developed and equipped with innovative technology to enable technically sophisticated measurement of absorption. iCheck Chroma displays vitamin A concentration in mg RE/kg.

iCheck Chroma is equipped with the latest light emit-ting diode (LED) technology. Using LEDs as the light source eliminates all known problems with halogen lamps used in other readers. Modern LEDs emit particularly stable light and have very low energy consumption. Furthermore, each iCheck Chroma is calibrated at the production site in Germany with vitamin A standards according to quality manage-ment system. This eliminates the need for the user to calibrate the device. Based on the half life of the LED, approximately 10 million measurements can be performed reliably on iCheck. iCheck Chroma reagent vial is a 2-in-1 disposable reaction vial and cuvette. iCheck Chroma reagent vials contain a certified volume of reagents to determine vitamin A in the sample. One iCheck Chroma test kit consists of 100 reagent vials. No additional chemicals or equipment are required.

No, there is no need to calibrate iCheck Chroma, because the device is calibrated during the manufac-turing process and calibration is programmed into the software.

    1. iCheck Chroma variation for repeated measure-ments of the same sample is below 10%. Therefore 10% deviation from expected value is acceptable.

    2. The oil sample was not well mixed before taking up with the syringe. Mix the oil sample well before tak-ing up into the syringe.

    3. No device warm-up was carried out.

    4. Incorrect execution of the test: It is very important to mix the vial at exactly 0. Insert the vial immediately after mixing. Maximum waiting time allowed between mixing and inserting into iCheck Chroma is 2 seconds.

Do I need to calibrate iCheck Chroma?

The result I received for a sample is higher/lower than expected. What might be the reason for this?

    Frequently AskedQuestions

    5. Incorrect volume:Make sure that exactly 500 L (0.5 mL) of sample is injected into iCheck Chroma reagent vials.

    6. Incorrect injection:There has to be a distinct interface between oil sam-ple and reagents in the vial before mixing.

    7. The operator was not well trained. Contact BioAnalyt to obtain training and iCheck certification.

    1. Unclean vial surface: Make sure the reagent vial you are measuring is ab-solutely clean and does not have any fingerprints on it. If not, wipe the vial with a paper tissue (optional: wet the tissue with alcohol to improve the cleaning).

    2. Sunlight: Do not measure in the direct sunlight.

What might interfere with the measurement procedure?

    Measurement

  • 21

iCheck reagent vials must be stored upright at room temperature and protected from direct sunlight.

    1. It is recommended to measure in the range of 20 - 30 C (68 - 86 Fahrenheit). Do not use iCheck at tem-peratures above 40 C.

    2. It is further recommended to store the iCheck and the iCheck reagent vials at least two hours before starting the measurement in the room in which the measurement will be performed. This procedure ensures that both, the vials and the device have the same temperature.

    3. Humidity does not aect iCheck measurements.

For selecting a new batch press the right arrow key. The batch function is used to group samples, e.g. samples from 1 day or 1 region can be measured in batch 1. If you proceed to measure the samples of a dierent day or region, select a new batch (i.e. 2).

How should I store the reagent vials?

Do temperature or humidity inuence the iCheck measurements?

What is a batch and how can I select a new batch?
