CoolLED

CoolLED is a UK-based specialist LED technology manufacturer

It is owned by Custom Interconnect Ltd. (CIL), an established electronics manufacturing company

CoolLED’s Life Sciences & Analytical division supplies the pE family of LED excitation products

CoolLED builds its LED products using the bare semiconductor LED die to achieve the highest possible performance.

CoolLED is not restricted to commercially available packaged LED products which means it can offer a greater range of wavelengths, stability and intensity

LED excitation

Narrow waveband Stable

Better results from less damaged samples

Reliable comparisons of quantitative measurements

Active-cooling of LEDs using a Peltier gives

best intensity and stability

Repeatable, reliable, and comparable results

Not exposing samples to unwanted wavelengths

e.g. UV

Less photo-bleaching

LED Excitation

LEDs are Controllable

Instant “On/Off”0-100% Intensity

in 1% steps

No warm up or cool down times

Optomise intensity independently foreach fluorophore

Intensities can be pre-set for a given test

Repeatable and consistent results

Switching does not affect LED lifetime

Fast switching between wavelengths

~300 uSecs with TTL

Perfect for pre-aligning samples on a Confocal

LEDs are controllable

Costs & Safety

Long lifetime measured in tens of thousands

of “On” hours

LEDs do not contain Mercury (Hg) which

is hazardous

No replacement lamps to purchase

No risk of explosion or H&S risk from Mercury vapour

International regulations are removing Mercury

from most products

No Hg disposal issues. No H&S regulations to

follow for operator

Switching does not affect LED lifetime

No need to re-align when replacing lamps

Costs & Safety

Better Results

Quantitative Live Cell

Meaningful comparisons can be made between measurements made

at different times

No vibration, no delay, no transfer of heat or

electromagnetic radiationto sample

A growing area for Fluorescence microscopy

as results can be more meaningful on living cells

Optimised intensity for each fluorophore. Dual staining applications do not require

an excitation filter wheel

CoolLED excitation is stable and repeatable due to active-cooling

Perfect for Time Lapse

Better Results with LEDs

Mercury - Metal Halide (contains Mercury) - Xenon

All suffer from the same drawbacks:-

• Short Lifetime

• Warm-up/cool-down period

Decreasing Intensity

• Use hazardous materials

• H&S / Disposal issues

• On-going cost of lamps

• Lamps need re-aligned

• Unstable

Traditional Lamps

Mercury LampsStrength Counter Argument

Broadband excitation Most needs to be filtered out

More intense Only at some wavelengths

Unwanted intensity can damage samples

Mercury Lamps

12

Weakness Awareness Proposal

Warm-up / Cool-down period LEDs are almost instant

Declining intensity through life LED intensity remains for tens of thousands of hours

Lamps need to be replaced regularly

Virtually no running costs with LED systems

Lamps need re-aligned regularly CoolLED is aligned once-ever at installation

Lamps can explode Although rare, it is a serious H&S risk

Mercury Lamps

Strength Counter Argument

Broadband excitation Most needs to be filtered out

More intense Only at some wavelengths

Unwanted intensity can damage samples

Longer life than Mercury Still short by LED standards

Still declines during life

Strength Counter Argument

Integrated with Axiovision Adds to investment cost as user requires latest version

Simultaneous excitation Often requested but most cameras are monochrome so fast switching is actually what is required. CoolLED is developing simultaneous in the future

Weakness Awareness Proposal

Expensive CoolLED pE-2 is ~€6K* List with 4λ * Based on current exchange from £5375

Intensity in Green is an issues IMP Vienna (Zeiss lab) reported that CoolLED is 4-5x intense in this region. Purchased 12 units

Only integrated with Axiovision Excludes users using NIS Elements, MetaMorph, MicroManager, etc.

Latest version of Axiovision required and adds to cost

Only 10 wavelengths available CoolLED has 18 and will develop more

Thor Labs Single wavelength

Lower intensity

No integration

Cairn Research Less wavelength options

Less intense

Fraen Less intense

Transmitted light option

Other LED sources

All other LED product suppliers are limited to using commercially available LED packaged parts which are not actively-cooled to achieve

CoolLED’s extra intensity and stability
