The conventional dosage form provide drug release for

immediately and it cause fluctuation of drug level In blood.

Therefore to maintain the drug concentration in blood

within therapeutic effective range need noval drug delivery

systems.

WHY DO WE NEED NOVEL DRUGS

TYPES OF NOVAL DRUG DELIVERY SYSTEM

NOVAL DRUG

DELIVERY

Oral drug delivery

Inhalation / pulmonary

drug delivery system

Tans dermal drug delivery

systemSpecializati

on drug delivery system

Injection based

delivery system

GI specific delivery

ORAL DRUG DELIVERY SYSTEM

Formulation range from simple tablets to newest

modified tablets

Involves use of various forms of hydrogels, metrics

andpolymers.

Modification in oral NDDS based on:

Dissolution

Osmotic pressure

Microsphere based technology

Novel Packaging

•Dissolution and diffusion controlled system:

•Osmotic pressure controlled system

(Nefidipine)

VARIOUS TECHNOLOGY TO IMPROVE

BIOAVAILABILITY:

Micro cap delivery system(bio vail canada)

Micro pump delivery system( flamel tech, France)

Zeros tablet technology (switzerland)

Ceform micro sphere technology (fuisz technology ltd. , USA)

DPHS ( Delayed Pulsetile Hydrogel System): Andrex

pharmaceuticals

Dual release drug absorption system ( Elan Corporation)

Programmable Oral Drug Absorption System (elan corporation)

GI Specific drug release System

For sustain release of drug in GIT

Various approach to prolong GI residence time

-GAS generating system

-Super porous hydrogel

-Mucoadhesive systems

-Ex: Buccasartan

(buccal tablet , used for treatment of nausea and vomitting)

-Colon Targeted Drug delivery:

-in treatment of Colon

- hydrogels, prodrugs and coated dosage forms

basically preferred

INJECTABLE DRUG DELIVERY SYSTEM

Insulin Delivery : Insulin Pens Insulin Pumps

• Bone Marrow Injections (in chronic myocardial intra myocardial

injections of Autologous bone marrow cell)

Inhalational Drug Delivery System

• A bright Prospect for site specific Delivery of biotechnological

Products such as insulin and other hormones.

• Attractive For delicate molecule allowing systemic circulation

without significant degradation

Metered dose inhaler

Metered dose inhaler. Dry powder inhaler

Inhalation solution & suspension (

nebulizer)

Inhalation Nasal Spray

Transdermal Delivery System

•Provides a leading edge over injectables and oral

routes by increasing.

•Controlled , constant administration of the drug , allows

continuous input of drugs with short biological half life.

•Transdermal Patches

•Micro needle patches : for improving the absorption of

drugs.

Various Technology Used

Coated Implantable Drug Delivery System

• Permits Localized Drug Delivery from implant.

• Best for prolonged duration

EXAMPLE :

Hormonal delivery to endometrium from intrauterine

device, sirolimus coated stents in coronary arteries.

Targeted Drug Delivery

Monoclonal Antibodies: Bind with cancer specific antigen and

induce immunological response to cancer cell.

Modify for delivery toxin, radioisotops, cytokine

or other active conjugatates.

LIPOSOMAL DRUG DELIVERY SYSTEM

Pre-clinical and clinical liposomal packed drug exhibit reduced

toxicities with enhanced efficacy,

Drug accumulation at diseased sites and reduced distribution

example: Amphotericin B antibiotics

Nano Particle Based Delivery system

• By using nana particle drug can be used at the precise location,

which would make the drug more effective.

• It reduces the toxicities , greater safety and biocompatibility.

EX: carbon nano tube: treatment of Bronchial Asthma, lung

toxicities

•Gold nano particle :used in

chemotherapy n

free radical generation

Application of Nanotechnology:

•Promising tool for advancement of drug delivery

•Nano shells that concentrate the heat from infrared light to

destroy cancer cell with minimal damage to surrounding healthy

cells

•Nanotubes used in broken tubes to provide a structure for new

bone material to grow.

•Nano particle in pulmonary drug delivery system.

Nano erythrosomes

Erythrocytes based new carrier.

Administered via iv or intra-arterial.

Prepared by extrusions of erythrocyte

ghost to produce small vesicle.

Genetic Transfer system

Gene transfer by several nano formulations

are still under formulations.

Tow methods- viral or non viral gene transfer.

Currently very few are in phase 3 trial.
