SUMMARY
Herpes Simplex virus type 1 thymidine kinase (HSV 1 TK) catalyzes the
phosphorylation of thymidine (dT) in the presence of adenosine triphosphate(ATP) and magnesium ions. HSV 1 TK exhibits a broad Substrate diversity. It
phosphorylates not only pyrimidine analogs (e.g. BVdU and IdU) but also guanineand adenine analogs. It furthermore accepts bases carrying acyclic or carbocyclicsugar moiety Substitutes. In contrast, human cellular TK exhibits a rather highSubstrate specificity and accepts only pyrimidine Substrate analogs and restricted
changes in the sugar moiety. This is the crucial molecular basis for therapeuticapplications. Interestingly, HSV 1 TK is able to phosphorylate thymidinemonophosphate, a feature which raises questions on evolutionary relationships.TK plays an important role in the reactivation of the herpesviruses from latencyand might therefore be a valuable target to prevent recurrent virus infections, the
major obstacle of herpesvirusinfection. TK established itself as major tool in many
applications such as gene therapeutical treatment of cancer and AIDS, as control
system in allogeneic bone marrow transplantation and AIDS Vaccine, as
expression reporter gene and is an important target in antiviral therapy.The main aspect of this work is the exploration of the mechanisms that
contribute to the unique Substrate acceptance by the molecular characterization of
TK - ligand interactions with the aid of site-directed mutagenesis experimentsbased on the crystal structure. Insights into the structural requirements for TK-
substrate interaction provide and widen the knowledge base which facilitates and
supports rational ligand design suitable for various eventualapplications.Several site-directed mutagenesistechniques (based on Single stranded DNA
or Polymerase chain reaction (PCR) methods) were tested with the HSV 1 TK
gene which exhibits a rather high G/C content of up to 68%. Since its Singlestranded DNA seems to have a high propensity to form stable secondarystruetures,the best mutagenesisresults were achieved using PCR. A three and a
four primer PCR method were established and employed depending on the
location of the desired mutation. The sequencing technique was reorganized to
automatedsequencingin order to increase the scientific Output.

Summary
The wild-type and mutantenzymes of HSV 1 TK were expressed in E. coli BL 21
as thrombin- or PreScission protease-cleavableglutathione S-transferase-fusion
proteins and were purified to homogeneityin a single-step procedure. The mutant
enzymeswere kinetically characterized in a partially truncated form in the case of
thrombin cleavage, as a compromise for stability reasons. Full-Iength TK was
derived from PreScission protease cleavage and used for crystallization. For
activity Screenings and ATP kinetics, a coupled UV-spectrophotometric assayinvolving pyruvate kinase and lactate dehydrogenase was applied. For more
accurate analysis of Substrate conversion, a HPLC assaywas established and the
Standard nucleoside kinase assay, the DEAE-paper method, was further
optimizedand used for determinationof the kinetical parameters.Mechanismsof Substrate binding by HSV 1 TK were studied by means of site-
directed mutagenesis guided by theoretical calculations and sequence
comparisonand combined with isothermal calorimetricmeasurements. The results
show the link between the exceptionally broad Substrate diversity of HSV 1 TK
and the presenceof structural features such as the residue triad H58/M128/Y172.The mutation of Met 128 into a Phe as well as the double mutant M128F Y172F
result in mutants that have lost their activity. However, by exchanging His to form
the triple mutant H58L.M128F/Y172Fthe enzyme regains activity. Strikingly, this
triple mutant becomes resistant towards acyclovir. Furthermore, the importance of
Glu 225 of the flexible LID region for the catalytic reaction is outlined. These data
give new insights in understanding mechanisms ruling Substrate diversity.Mutations previously identified as relevant in drug resistance were studied in an
attempt to further understand their role in these processes. Four amino acid
positions are described (Q125, A168, R176 and C336), each of which conferdrugresistance when mutated, however, the molecular mechanismsare considerablydifferent in each case. The results reveal three main mechanisms ruling drugresistance. Two of them , the loss of electrostatic interactionsand the alteration of
steric accessibility are "rational" from a structural point of view. Whereas the third
one, the modification of the 3D conformation is more surprising and is caused bylong ränge effects of a distant mutation.
Additional mutants (Y101H, Q125S, M128K) that provide valuable insight in
enzyme-ligand interaction, enzyme rearrangement and in the subtle balance in

electrostatics are presented. Substrate diversity was further investigated byanalyzing the phosphorylation capability of HSV 1 TK and Varicella zoster TK
using a set of unique 9-hydroxypropyl-substituted nucleoside analogs combinedwith structure-activity studies.
An attempt in protein engineeringwas undertaken to screen the use of TK as a
phosphorylationtool ultimately leading to non Standard DNA building blocks ableto establish base pairing. Thereto, a series of mutants was established in an
essay renderingthe electrostatics in the active site more favorable for Compoundconversion. Furthermore, with the mutants A168VL169Mand G129AL169M, we
were able to design mutants that exhibit an altered Substrate specificity. Moreover,these effects are caused by mutations distantfrom the active site.
In conclusion, a combination of practical work comprising site-directed
mutagenesisand subsequent characterization of the emerging mutant enzymesand theoretical calculations based on the crystal structure has been used to
deepen the knowledgecrucial for both the developmentof new antiviral drugs and
engineering of mutant TKs apt to accept novel Substrate analogs for genetherapeutic approaches.

Thymidinkinasen katalysieren die Phosphorylierung von Thymidin (dT) zu
Thymidinmonophosphat unter Dephosphorylierung von Adenosintriphosphat(ATP) in Anwesenheit von Magnesiumionen. Im Gegensatz zum humanenzellulären Enzym besitzt die Thymidinkinase des Herpes Simplex Virus Typ 1
(HSV 1 TK) die Eigenschaft, nicht nur das natürliche Substrat Thymidin, sondernauch eine ganze Reihe weiterer Substrate wie zum Beispiel Guanosin- und
Adenosinanaloga zu phosphorylieren. Dieser Unterschied bezüglich der
Substratspezifität ermöglicht eine selektiv antivirale Therapie. HerpesviraleTK ist
sogar in der Lage, ihr Katalyseprodukt, Thymidinmonophosphat, umzusetzen,eine Besonderheit, die Fragen bezüglich der evolutionären Abstammung derHSV 1 TK aufwirft. Darüberhinaus scheint sie bei der Reaktivierungder Viren aus
der Latenzphase in neurononalen Ganglien eine wichtige Rolle zu spielen. Einepermanente Hemmungder viralen TK wäre also eine gute Präventivmassnahme,um dieses zentrale Problem einer Infektion mit Herpesviren zu lösen.
Das Hauptanliegen dieser Arbeit bestand in der Charakterisierung der
Wechselwirkungenvon TK mit ihren Liganden auf molekularer Ebene, basierendauf der Kristallstruktur des Proteins. Dazu wurden gezielt einzelne Positionen der
Aminosäuresequenz verändert, um die grundlegenden Mechanismen der
Akzeptanz verschiedenster Substrate zu verstehen. Strukturelle Einblicke in die
Enzym-Substrat-Interaktiontragen zur Erweiterung des Wissens, das für den
rationalen Entwurf von neuen TK-Substraten für biomedizinischeZwecke nötig ist,bei. Neben der antiviralen Therapie hat sich vor allem der Einsatz von TK zur
gentherapeutischen Behandlung von Krebs und AIDS etabliert; ausserdem kannTK als „Notbremse" bei Komplikationen von Knochenmarktransplantationen von
Fremdspendern und bei Gefährdung durch eine retrovirale AIDS Impfungangewendet werden.
Mehrere Technikenzur zielgerichtetenMutagenese (basierendauf dem Einsatzvon einzelsträngiger DNS oder der Polymerase Kettenreaktion) wurden auf ihre
Tauglichkeit zur Anwendung am TK-Gen hin getestet. Dieses weist einen sehrhohen Gehalt an Guanin und Cytosin auf und neigt aus diesem Grund zur
Ausformung stabiler Sekundärstrukturen. Wir etablierten je nach Lage derauszutauschenden Aminosäure zwei unterschiedliche Mutagenese-Methodenunter Verwendungvon drei bzw. vier Oligonukleotiden und stellten von manueller

auf automatische Sequenzierungder mutierten Gene um. Wildtyp und Mutanten
wurden in Colibakterien als Fusionsproteine, die mit Thrombin oder PreScissionProtease spaltbar sind, exprimiert.Nach einer einstufigen Reinigung erfolgte eine
Charakterisierungder Enzyme mittels zunächst UV-spektrophotometrischer und
hochdruckchromatographischer Verfahren. Im Anschluss wurde die
Kinaseaktivität mit der DEAE-Papiermethode,exakt bestimmt.Die Kombination von zielgerichteter Mutagenese und kalorimetrischen
Messungen ermöglichte, gestützt auf theoretische Berechnungen und
Sequenzvergleiche zwischen verschiedenen TK-Species, eine detaillierte
Untersuchung der Substratbindung. Dabei wurde ein Zusammenhangzwischender breiten Substratakzeptanz der TK und der Aminosäure-Triade
H58/M128/Y172offenkundig. Während die Mutanten M128F und M128F/Y172F
inaktiv waren, erlangte die Tripelmutante H58L/M128F/Y172Fdie Aktivität zurück
und erwies sich ausserdem gegenüber Acyclovir als resistent. Zusätzlichuntersuchten wir mit der Mutante E225L den Einfluss der beweglichen „LID"Domäne auf die Katalyse der TK.
TK-Mutanten, die schon früher als relevant für Arzneimittel Resistenz erkannt
worden waren, wurden in struktureller und teilweise auch kinetischer Hinsicht
untersucht. Die Positionen Q125, A168, R176 und C336, rufen Resistenz hervor,wenn sie verändert werden. Dabei unterscheiden sich die molekularen
Mechanismen aber beträchtlich. Verantwortlich für Resistenzentwicklung sind
einerseits der Verlust an Wasserstoffbrücken-Wechselwirkungen oder sterisch
bedingte Änderungen der Zugänglichkeit der Bindungstasche für das Substrat.
Andererseits kann auch die Veränderung der dreidimensionalen StrukturResistenz bewirken.
Weitere Mutanten (Y101H, Q125S, M128K) sind vorgestellt, die wertvolle
Aussagekraft bezüglich der Enzym-Ligand-Interaktionund Elektrostatikinnerhalb
der Bindungstasche besitzen und deshalb das Wissen über den
massgeschneiderten Entwurf von Proteinen für spezielle Verwendungszweckeerweitern und vertiefen.
Die Substrat-Diversität sowohl der HSV 1 TK als auch der Varizella Zoster TK
analysierten wir anhand einer Reihe von verschiedenen Basen, die an der
Position 9 mit einem Hydroxypropylrestsubstituiert worden waren. Ein Versuch in
gezieltem „Protein-Entwurf" wurde unternommen, um die TK derart zu verändern,

dass sie neue DNS-Bausteine phosphorylieren kann. Dazu versuchten wir, eineReihe von Mutanten mit optimal auf die Basen ausgerichteter Elektrostatikinnerhalb der Bindungstasche herzustellen.
Strukturelle Auswirkungen des Austauschs von Aminosäuren, die nicht in
unmittelbarer Nähe der Bindungstasche liegen, wurden richtig interpretiert und mit
der Gestaltung der MutantenA168VL169Mund G129AL169Mbestätigt.Insgesamtkönnen die Resultate dieser Arbeit, die auf dem Zusammenspiel von
praktischen Experimenten und theoretischen Berechnungen beruht, hilfreich fürdie Entwicklung von neuen Arzneistoffen mit antiviraler Wirkung sein und die
Konstruktion von genetisch veränderterTK ermöglichen, die durch ihre neuartigeSubstratakzeptanz für gentherapeutische Behandlungsansätze besonders
geeignet ist. Gleichzeitigtragen die Resultate zu einem besseren Verständnis der
physiologischenund pathologischen Rolle derTK bei.
