Pregnancy Increases Oral Clearance of Metoprolol

To assess the inlluence at pregnancy on the kinetics of metoprolol, 5 women with pregnancy-induced hypertension were given single oral (l00mg) and IV (1 0mg) doses during the third trimester and 3-6 months after delivery.

During pregnancy the pharmacokinetics of intravenously administered meloptolol were not significantly altered. However. the AUC increased from 998 nmol · hour/l during pregnancy, to 3562 nmol· hour!L after delivery. The apparent oral clearance feU from 9 .56 to 1.71 lImin, and the lolal bioavai1ability increased flom 0.21 during pregnancy to' 0.42 after delivery. Thus, the apparent oral clearance at metoprolol is increased during pregnancy, and plasma levels may need to be monitored in patients with pregnancy-induced hypertension. HOgs/edt. S.: ~g. a : Peng.
